目的:通过检测大麻素(CB)1、2型受体(CB-1R、CB-2R)在大鼠累积损伤性椎间盘炎症反应过程中的表达来探讨其中大麻素的参与过程及作用机制。方法清洁级Wistar大鼠随机分成实验组与对照组,根据Jill A.Ulrich制作方法准备实验组,以切皮后即缝合为对照组,在术后第3、7、10 d,实验组与对照组每次各取6只,取出相应椎间盘,进行形态学观察,应用RealTime-PCR技术检测CB-1R、CB-2R的表达水平;利用Western Blot方法检测CB-2R蛋白含量。结果与对照组相比,实验组10 d可见刺伤所致纤维环断裂,髓核内出血。在椎间盘累积性损伤后,CB-1R、CB-2R呈时间依赖性增高趋势。3、7、10 d组CB-2R蛋白表达与对照组相比明显增高( P<0.01)。结论在骨骼椎间盘反复损伤及其后炎症反应阶段,CB类物质参与了炎症反应的发展过程,从而抑制骨骼椎间盘的退变。

목적:통과검측대마소(CB)1、2형수체(CB-1R、CB-2R)재대서루적손상성추간반염증반응과정중적표체래탐토기중대마소적삼여과정급작용궤제。방법청길급Wistar대서수궤분성실험조여대조조,근거Jill A.Ulrich제작방법준비실험조,이절피후즉봉합위대조조,재술후제3、7、10 d,실험조여대조조매차각취6지,취출상응추간반,진행형태학관찰,응용RealTime-PCR기술검측CB-1R、CB-2R적표체수평;이용Western Blot방법검측CB-2R단백함량。결과여대조조상비,실험조10 d가견자상소치섬유배단렬,수핵내출혈。재추간반루적성손상후,CB-1R、CB-2R정시간의뢰성증고추세。3、7、10 d조CB-2R단백표체여대조조상비명현증고( P<0.01)。결론재골격추간반반복손상급기후염증반응계단,CB류물질삼여료염증반응적발전과정,종이억제골격추간반적퇴변。
Objective To reveal the function of CB during disc inflammation due to cumulative damage by detecting CB-1R and CB-2R.Methods Wistar rats were randomly divided into experiment group and control group.The experiment group was prepared according to Jill A Ulrich method.Control group was prepared by skin cutting and suturing.Six rats were taken from each group on 3rd day, 7th day, and 10th day after the operation.The intervertebral discs were collected.Morphology was observed. Expression of CB-1R and CB-2R was detected using real-time PCR.CB-2R protein content was examined using Western blotting. Results Compared with the control group, the annulus rupture and nucleus bleeding were observed in experiment group in 10 days.The expression levels of CB receptor increased significantly in a time-dependent manner.CB-2R protein expression increased more significantly at the 3rd day, 7th day, and 10th day in experiment group than that in control group (P<0.01).Conclusion CB is involved in the regulation of the rat bone and disc inflammatory reaction due to the cumulative damage.
