目的:研究贲门腺癌患者癌组织 RKIP 基因甲基化和 mRNA 表达情况及其临床意义。方法选择2008—2013年河北工程大学附属医院收治的贲门腺癌患者160例,采用以Taq Man探针为基础的实时定量聚合酶链反应(PCR)和反转录-聚合酶链反应( RT -PCR)检测贲门腺癌患者癌组织及癌旁正常组织 RKIP 基因甲基化和mRNA表达情况。结果贲门腺癌患者癌组织RKIP基因甲基化发生率为68.1%(109/160),高于癌旁正常组织的35.6%(57/160)(χ2=33.843,P=0.000)。不同年龄、性别、临床分期贲门腺癌患者RKIP基因甲基化发生率比较,差异无统计学意义(P>0.05);不同病理分级及有无淋巴结转移贲门腺癌患者癌组织RKIP基因甲基化发生率比较,差异有统计学意义( P<0.05)。贲门腺癌患者癌组织RKIP基因mRNA阳性表达率为48.1%,低于癌旁正常组织的71.9%(χ2=18.802,P<0.01)。癌组织RKIP基因甲基化患者RKIP基因mRNA阳性表达率低于癌组织RKIP基因非甲基化患者(χ2=6.410,P<0.01)。结论 RKIP基因甲基化导致该基因mRAN表达缺失可能是贲门腺癌的发病机制之一,且RKIP基因的高甲基化与贲门腺癌患者病理分级和淋巴结转移有关。

목적:연구분문선암환자암조직 RKIP 기인갑기화화 mRNA 표체정황급기림상의의。방법선택2008—2013년하북공정대학부속의원수치적분문선암환자160례,채용이Taq Man탐침위기출적실시정량취합매련반응(PCR)화반전록-취합매련반응( RT -PCR)검측분문선암환자암조직급암방정상조직 RKIP 기인갑기화화mRNA표체정황。결과분문선암환자암조직RKIP기인갑기화발생솔위68.1%(109/160),고우암방정상조직적35.6%(57/160)(χ2=33.843,P=0.000)。불동년령、성별、림상분기분문선암환자RKIP기인갑기화발생솔비교,차이무통계학의의(P>0.05);불동병리분급급유무림파결전이분문선암환자암조직RKIP기인갑기화발생솔비교,차이유통계학의의( P<0.05)。분문선암환자암조직RKIP기인mRNA양성표체솔위48.1%,저우암방정상조직적71.9%(χ2=18.802,P<0.01)。암조직RKIP기인갑기화환자RKIP기인mRNA양성표체솔저우암조직RKIP기인비갑기화환자(χ2=6.410,P<0.01)。결론 RKIP기인갑기화도치해기인mRAN표체결실가능시분문선암적발병궤제지일,차RKIP기인적고갑기화여분문선암환자병리분급화림파결전이유관。
Objective To observe the methylation and mRNA expression of RKIP gene in cancerous tissues of patients with cardia adenocarcinoma,to explore its clinical significances. Methods From 2008 to 2013,a total of 160 patients with cardia adenocarcinoma were selected in the Affiliated Hospital of Hebei Engineering University,and Taq Man probe-based real-time quantitative PCR and RT-PCR were used to detect methylation and mRNA expression of RKIP gene in cancerous tissues and corresponding normal tissues. Results The methylation rate of cancerous tissues was 68. 1%, was higher than that of corresponding normal tissues of 35. 6%(χ2 =33. 843,P=0. 000). No statistically significant differences of methylation rate was found in patients with different ages,gender or clinical stages(P>0. 05);but there was statistically significant differences of methylation rate in patients with different pathological gradings and with or without lymphatic metastasis ( P <0. 05 ) . The positive expression rate of RKIP gene mRNA of cancerous tissues was 48. 1%,was lower than that of corresponding normal tissues of 71. 9%(χ2 =18. 802,P<0. 01). The positive expression rate of RKIP gene mRNA of patients with RKIP gene methylation was lower than that of patients without RKIP gene methylation (χ2 = 6. 410, P < 0. 01 ). Conclusion The incomplete expression of RKIP gene mRNA caused by RKIP gene methylation may play a role in the mechanism of cardia adenocarcinoma, and the methylation of RKIP gene is correlated with pathological gradings and lymphatic metastasis.
