目的:分析胃癌组织中亲环素 A(CypA)表达与肿瘤细胞体外化疗药物敏感性及耐药相关蛋白的关系,并探讨其意义。方法磺酰罗丹明 B(Sulforhodamine B,SRB)法检测94例胃癌组织对氟尿嘧啶(5-FU)、奥沙利铂(L-OHP)、阿霉素(ADR)的体外化疗药物敏感性;对相应石蜡标本进行免疫组化染色,检测 CypA 蛋白及耐药相关蛋白 P-糖蛋白(P-glycoprotein,P-gp)、谷胱甘肽-S-转移酶-π(Glutathione S-transferase-π,GST-π)、x-IAP、Survivin 的表达。结果 CypA、P-gp、GST-π、x-IAP、Survivin 阳性表达率分别为72.3%(68/94)、90.4%(85/94)、62.8%(59/94)、66.0%(62/94)、76.6%(72/94),均明显高于癌旁组织(均 P <0.05)。5-FU、L-OHP 对 CypA 强表达的肿瘤细胞抑制率低于弱表达者(均 P <0.05),而 CypA 的表达情况与 ADR 对肿瘤细胞的抑制率无明显关系(P >0.05)。相关分析显示,CypA 蛋白与 P-gp、x-IAP、Survivin 均存在正相关关系(均 P <0.05)。结论胃癌细胞对5-FU、L-OHP 药物敏感性与 CypA 蛋白表达有关,该作用可能是 CypA 调节部分耐药相关蛋白实现的。

목적:분석위암조직중친배소 A(CypA)표체여종류세포체외화료약물민감성급내약상관단백적관계,병탐토기의의。방법광선라단명 B(Sulforhodamine B,SRB)법검측94례위암조직대불뇨밀정(5-FU)、오사리박(L-OHP)、아매소(ADR)적체외화료약물민감성;대상응석사표본진행면역조화염색,검측 CypA 단백급내약상관단백 P-당단백(P-glycoprotein,P-gp)、곡광감태-S-전이매-π(Glutathione S-transferase-π,GST-π)、x-IAP、Survivin 적표체。결과 CypA、P-gp、GST-π、x-IAP、Survivin 양성표체솔분별위72.3%(68/94)、90.4%(85/94)、62.8%(59/94)、66.0%(62/94)、76.6%(72/94),균명현고우암방조직(균 P <0.05)。5-FU、L-OHP 대 CypA 강표체적종류세포억제솔저우약표체자(균 P <0.05),이 CypA 적표체정황여 ADR 대종류세포적억제솔무명현관계(P >0.05)。상관분석현시,CypA 단백여 P-gp、x-IAP、Survivin 균존재정상관관계(균 P <0.05)。결론위암세포대5-FU、L-OHP 약물민감성여 CypA 단백표체유관,해작용가능시 CypA 조절부분내약상관단백실현적。
Objective To analyze the relationship between chemosensitivities of gastric cancer cell,drug-resistant related protein and protein expression of cyclophilin A (CypA)in vitro,and to explore its significance. Methods In 94 tissues of gastric cancer,the chemosensitivities of tumor cell to 5-FU,L-OHP,ADR in vitro were tested by SRB assay, and expression of CypA, P-gp, GST-π, x-IAP and survivin were determined by immunohistochemical method.Results The positive expression of CypA,P-gp,GST-π,x-IAP,survivin were 72.3%(68/94),90.4%(85/94),62.8%(59/94),66.0%(62/94)and 76.6%(72/94),respectively,and protein expression of CypA,P-gp,GST-π,x-IAP,survivin were significantly higher in tumor cell than in adjacent non-tumor cell (P <0.05).The inhibition rates of 5-Fu,L-OHP to tumor cell were significantly lower in high expression of CypA group than in low expression of group (P <0.05),whereas there were no significant difference between inhibition rates of ADR to tumor cell and expression of CypA (P >0.05).CypA protein was positively associated with P-gp,x-IAP and survivin (P <0.05).Conclusion The protein expression of CypA was associated with the chemosensitivities of 5-FU, L-OHP in gastric cancer cell,which may be implemented by CypA through regulating drug-resistant related proteins.
