目的:探讨芦黄颗粒抗动脉粥样硬化的作用及其部分机制。方法30只大白兔随机分为3组,即正常对照组(10只)、模型对照组(10只)、芦黄颗粒组(10只);正常对照组饲普通标准饲料,模型对照组、芦黄颗粒组先导管球囊损伤兔左侧颈总动脉然后喂饲高脂饲料;同时芦黄颗粒组每天灌胃芦黄颗粒液3ml/kg,而正常对照组、模型对照组灌胃等量生理水。实验满12周后停药禁食12h,耳缘静脉取血10ml用于做血脂、Hs-CRP检查;用10%乌拉坦耳缘静脉注射麻醉,分离颈总动脉,观察大体在左总动脉血管节段斑块病变情况;剪取尽可能长的颈总动脉血管节段均石蜡包埋,做连续切片,油红O染色。油红o染色后,标本平铺,使用数码相机拍照,用专业图像分析软件IPP 6.0测定颈动脉面积、粥样斑块面积,以及板块面积占颈动脉面积的百分比。左颈总动脉斑块组织,采用免疫组织化学法测定组织Hs-CRP含量。结果正常对照组与模型对照组血脂,血清Hs-CRP水平有显著性差异(P<0.01),3组血清及斑块组织Hs-CRP含量差异有显著性(P<0.01);模型对照组、芦黄颗粒组血清及斑块组织hs-CRP含量显著高于正常对照组(P<0.01),芦黄颗粒组血清及斑块组织hs-CRP含量显著低于模型对照组(P<0.01)。肉眼可见正常对照组内膜光滑,无斑块明显形成;模型对照组血管表面粗糙,内膜面散在不规则表明隆突斑块,颜色为瓷白色;芦黄颗粒组血管内膜隆突斑块减少,颜色浅黄。与正常对照组相比,芦黄颗粒组斑块面积比无差异,P>0.05;与模型对照组相比,芦黄颗粒组斑块面积比小,P<0.05。结论芦黄颗粒有抗动脉粥样硬化的作用,其参与机制可能为调脂,抗炎症反应。

목적:탐토호황과립항동맥죽양경화적작용급기부분궤제。방법30지대백토수궤분위3조,즉정상대조조(10지)、모형대조조(10지)、호황과립조(10지);정상대조조사보통표준사료,모형대조조、호황과립조선도관구낭손상토좌측경총동맥연후위사고지사료;동시호황과립조매천관위호황과립액3ml/kg,이정상대조조、모형대조조관위등량생리수。실험만12주후정약금식12h,이연정맥취혈10ml용우주혈지、Hs-CRP검사;용10%오랍탄이연정맥주사마취,분리경총동맥,관찰대체재좌총동맥혈관절단반괴병변정황;전취진가능장적경총동맥혈관절단균석사포매,주련속절편,유홍O염색。유홍o염색후,표본평포,사용수마상궤박조,용전업도상분석연건IPP 6.0측정경동맥면적、죽양반괴면적,이급판괴면적점경동맥면적적백분비。좌경총동맥반괴조직,채용면역조직화학법측정조직Hs-CRP함량。결과정상대조조여모형대조조혈지,혈청Hs-CRP수평유현저성차이(P<0.01),3조혈청급반괴조직Hs-CRP함량차이유현저성(P<0.01);모형대조조、호황과립조혈청급반괴조직hs-CRP함량현저고우정상대조조(P<0.01),호황과립조혈청급반괴조직hs-CRP함량현저저우모형대조조(P<0.01)。육안가견정상대조조내막광활,무반괴명현형성;모형대조조혈관표면조조,내막면산재불규칙표명륭돌반괴,안색위자백색;호황과립조혈관내막륭돌반괴감소,안색천황。여정상대조조상비,호황과립조반괴면적비무차이,P>0.05;여모형대조조상비,호황과립조반괴면적비소,P<0.05。결론호황과립유항동맥죽양경화적작용,기삼여궤제가능위조지,항염증반응。
Objective To investigate effect and part of the mechanism of Luhuang granule on Anti-atherosclerosis in rabbits with atherosclerosis. Methods Forty white rabbits were randomly divided into three groups:normal control group (n=10),model group (n=10),Luhuang granule group (n=10). The normal control group have been feeded ordinary standard diet,the model group and the Luhuang granule group have been injured the left common carotid artery with balloon,and then feeded High-fat diet. At the same time,the Luhuang granule group have been feeded with Luhuang granule (3ml/kg,once a day) and the other two group have been feeded with the same capacity of NS. They have been Withdrawal fasted for 12 hours after 12 weeks. Then the contents of Hs-CRP and lipids have been detected by Ear vein blood about 10ml after With 10% urethane through ear vein injection of anesthesia,has been Clipped the left common carotid artery,Generally have been observed in the left main artery segment plaque lesions,have been Clipped it as long as possible segments.then they have been embedded in paraffin,been maken serial sections, been dyeinged by Oil red o staining,Tiled samples,been taken pictures Used a digital camera,been measured carotid area,plaque area and the percentage of the plate area occupied an area of the carotid artery with a professional image analysis software IPP6.0. The Left common carotid artery plaque tissue Hs-CRP content have been measured by Immunohistochemistry. Results The lipd levels difference is significantly between Normal control group from model group (P<0.01),and between Lu Huang particle group from the control group(P<0.01). The Hs-CRP levels in serum and tissue content difference were significant among the three groups (P<0.01). The Model group and Lu Huang particles groups Hs-CRP levels in serum and tissue content are significantly higher than the control group’(P<0.01). and the Huang particles groups Hs-CRP levels in serum and tissue content were significantly lower than the model group (P<0.01). Specimens eye view:It was intimal smooth and has No significant plaque on control group vascular surface;and it is intimal rough and has scattered in irregular patches bulge on model control groups vascular surface ,and its color was white porcelain;Lu Huang particle group intimal plaque was significantly less and color is light than the model groups (P<0. 05). Lu Huang particle group plaque area is no difference than the model control groups(P>0.05),and was smaller than the model groups (P <0.05). Conclusion Luhuang particles have anti-atherosclerotic effects,and it’part of the mechanism may be lipid transfer,be anti-inflammatory response.
