目的:建立一种操作容易、效果显著的大鼠动脉粥样硬化模型。方法8周龄Wistar大鼠分为高脂饲料组和基础饲料组,高脂饲料组大鼠给予按比例配制的高脂饲料,并辅以灌胃维生素D3,基础饲料组给予基础饲料,并灌胃生理盐水,喂养8周后观察大鼠血脂、血糖等生化指标、病理变化,并采用免疫组织化学法检测颈动脉中MMP-9的表达。结果经过8周高脂饲料喂养,高脂饲料组大鼠体重、血糖、甘油三酯、总胆固醇、低密度脂蛋白等生化指标明显高于基础饲料组,尤其是甘油三酯和总胆固醇水平;高脂饲料组大鼠出现大量钙化斑块,高脂饲料组大鼠颈动脉MMP-9表达增多,基础饲料组无明显变化。结论按照7%猪油、5%胆固醇、3%白砂糖、0.5%胆酸钠、0.2%丙硫氧嘧啶、84.3%基础饲料的比例配置高脂高糖饲料喂养大鼠,并辅以700000 U·kg-1的维生素D3进行灌胃,喂养8周可以成功建立大鼠动脉粥样硬化模型。

목적:건립일충조작용역、효과현저적대서동맥죽양경화모형。방법8주령Wistar대서분위고지사료조화기출사료조,고지사료조대서급여안비례배제적고지사료,병보이관위유생소D3,기출사료조급여기출사료,병관위생리염수,위양8주후관찰대서혈지、혈당등생화지표、병리변화,병채용면역조직화학법검측경동맥중MMP-9적표체。결과경과8주고지사료위양,고지사료조대서체중、혈당、감유삼지、총담고순、저밀도지단백등생화지표명현고우기출사료조,우기시감유삼지화총담고순수평;고지사료조대서출현대량개화반괴,고지사료조대서경동맥MMP-9표체증다,기출사료조무명현변화。결론안조7%저유、5%담고순、3%백사당、0.5%담산납、0.2%병류양밀정、84.3%기출사료적비례배치고지고당사료위양대서,병보이700000 U·kg-1적유생소D3진행관위,위양8주가이성공건립대서동맥죽양경화모형。
Objective To establish the model of atherosclerosis with easy operation,obvious effect in rats. Methods The 8 weeks old Wistar rats were divided into the high fat diet group and the basic diet group. The high diet rats were feed with high fat food which was prepared accooding to a certain proportion and Vitamin D3 by gavage. The basic diet rats were given ordinary rat food and physiological saline by gavage. After they were feed 8 weeks,biochemical parameters such as blood glucose and blood lipid and pathological changes were observed,the expression of MMP-9 was observede by immunohisto-chemical method. Results After the rats were feed with high diet food 8 weeks,the weight,the blood glucose,triglyceride,to-tal cholesterol,low density lipoprotein and other biochemical parameters of the high diet rats,especially triglyceride and the to-tal cholesterol,were higher than the basic diet rats'. There was a lot of calcified plaque was found in the high fat diet rats'ca-rotid artery,the expression of MMP-9 in the high fat diet rats increase,but there was no significant change in the basic diet rats. Conclusion The rats were feed with high fat food which include 7% lard oil,5% cholesterol,3%white sugar,0. 5% so-dium cholate,0. 2% propyl sulphur pyrimidine,84. 3% ordinary rat food,and 700 000 U·kg-1 Vitamin D3 by gavage. After 8 weeks,ideal rat model for the study of atherosclerosis can be established successfully.
