为筛选出最适蜜环菌栽培生产种的培养基,通过生产种培养基组分筛选试验,测量第20天和第40天时菌丝或菌索的生长速度及其胞外酶活性并进行比较。结果表明:在配方1(70%阔叶木屑,0%棉籽壳,17%麦麸,10%玉米面,1%蔗糖,0.3%磷酸二氢钾,0.2%硫酸镁,0.5%石灰,1%石膏,m水分∶m配料=1∶1.3)中生长速度最大且最先长满管,且菌丝洁白,与其余不同培养料配方之间生长速度差异显著,而且栽培种培养基含木屑越多,菌株活力越好,越不易形成老化的红褐色菌索;在不同培养料和不同生长阶段其木聚糖酶活性大于 CMC 酶和漆酶,漆酶活性最小,为十位级,其他2个酶为百位级;不同培养基中蜜环菌菌丝分泌的胞外酶活性具有显著性差异,但同种酶在不同培养料中酶活性的变化趋势一致。说明,配方1 可作为蜜环菌适宜的栽培种培养基;不同培养料对胞外酶活性变化趋势影响小,但对其活性影响较大。

In order to select the most suitable culture medium of A.mellea,the production of medium component was screened by measuring the A.hypha or bacteria growth speed and the extracellular enzyme activity,the authors compared between 20 and 40 days of hypha or rhizomorph of growth speed and extracellular enzyme activity.Results:Growth rate of the hyphae was the biggest and was the first full of tube and white in the formula(broadleaf sawdust 70% ,cotton seed shell 0,wheat bran 17%,corn flour 10%,sugar 1%,potassium dihydrogen phosphate 0.3%,magnesium sulfate 0.2%,lime 0.5%,gypsum 1%,mwater∶mmaterial=1∶1.3).The growth rate has obvious difference in different nutrient formula,and in culture medium,the more wood hyphae vitality the better,the more difficult to form a red brown rhizomorph;in different culture medium and different growth stages of the activity of xylanase and laccase enzyme is greater than CMC and laccase,the laccase activity is minimum for ten level,others for hundreds of level.Extracellular enzyme activity of A.mellea mycelia secretion of different medium has obvious difference,but the change trend in different culture medium in the same enzyme activity is consistent. Suggests that this formula can be used as armillaria suitable culture medium;different culture material has little effect on the enzyme activity changes in trend,but great influence on the activity of the extracellular enzyme.
