目的:通过观察预先使用杀菌性/通透性增加蛋白( BPI )的SD大鼠移植肝脏的转氨酶水平、TNF-α、白介素1(IL-1)水平的变化,探讨BPI预处理是否具有肝脏缺血再灌注损伤保护作用及其可能的相关机制。方法雄性SD大鼠96只,随机分为生理盐水对照组和 BPI组,Kamada’s袖套法经行肝移植。对照组切取供肝前10min 经腰静脉注射NS1.5mL;BPI组切取供肝前10min经腰静脉注射BPI(100μg/mL)1.5 mL。于再灌注后0h、3h及6h,酶联免疫吸附试验测定血清TNF-α及IL-1的含量;全自动血清自动生物化学仪检测血清转氨酶;光镜切片检测肝脏组织学。结果再灌注后0h,AST、ALT、TNF-α及IL-1含量两组间差异无统计学意义(P>0.05);再灌注后3、6h,NS组及BPI组各项指标水平逐渐升高,但BPI组各项指标水平明显低于NS组,差异有统计学意义(P<0.01)。结论 BPI对大鼠移植肝脏的缺血再灌注损伤有明显抑制作用。

목적:통과관찰예선사용살균성/통투성증가단백( BPI )적SD대서이식간장적전안매수평、TNF-α、백개소1(IL-1)수평적변화,탐토BPI예처리시부구유간장결혈재관주손상보호작용급기가능적상관궤제。방법웅성SD대서96지,수궤분위생리염수대조조화 BPI조,Kamada’s수투법경행간이식。대조조절취공간전10min 경요정맥주사NS1.5mL;BPI조절취공간전10min경요정맥주사BPI(100μg/mL)1.5 mL。우재관주후0h、3h급6h,매련면역흡부시험측정혈청TNF-α급IL-1적함량;전자동혈청자동생물화학의검측혈청전안매;광경절편검측간장조직학。결과재관주후0h,AST、ALT、TNF-α급IL-1함량량조간차이무통계학의의(P>0.05);재관주후3、6h,NS조급BPI조각항지표수평축점승고,단BPI조각항지표수평명현저우NS조,차이유통계학의의(P<0.01)。결론 BPI대대서이식간장적결혈재관주손상유명현억제작용。
Objective To observe the changes of TNF-α,interleukin-1, aspartate aminotransferase and alanine amin-otransferase from rat liver grafts after ischemia-reperfusion injury pretreated with bactericidal permeability increasing protein( BPI) and to explore the protective mechanisms of BPI pretreatment against hepatic I/R injury after liver transplantation. Methods Male Sprague-Dawley rats, weighing 180~220g, were divided into two groups in a randomized, blinded fashion. Normal saline group (NS group)and BPI pretreated group(BPI group). 10 min before we get the liver form donor, 1. 5ml normal saline or 1. 5ml BPI (100μg/ml)were injected in to two group form lumbar vein. After organ harvest, standardized liver transplantation was performed. At 0 h, 3 h and 6h after reperfusion, AST and ALT were detected by automatic serum automatic biochemistry instrument. TNF-αand IL-1 level in the plasma were detected by ELISA. Results After 0 hours of reperfusion, AST, ALT, TNF-alpha and IL-1 content of no significant difference between the two groups (P>0. 05); After 3 and 6 hours of reperfusion,NS group and BPI group each index level increased gradually, but the indicators of the level of BPI group was significantly lower than that of group NS (P<0. 01). Conclusion This data suggests that bactericidal permeability increasing protein can protect hepatic I/R injury after liver transplantation.
