通过筛选栽培花生间具有多态性的 SSR,分析总结多态性 SSR 的特征,旨在选择性利用 SSR 标记,推动栽培种花生遗传图谱构建、相关农艺性状的 QTL定位和分子标记辅助育种进程。以秦皇岛光秧、石腰豆、印度窄叶、抗019、四粒红和黑花生6个不同植物学类型的中国栽培种花生为材料,选用不同设计类型的1523个 SSR 标记筛选品种间多态性 SSR,并对其扩增片段长度、重复基序、重复次数等进行分析。结果表明,1523个标记中,筛选获得43对多态性 SSR标记,产生123个多态性标记位点,多态性标记数占2.8%。43对多态性 SSR 标记中,以 ARS ××的多态性最高,在6个品种中多态率达6.8%。统计各种质间具多态性的 SSR,其中石腰豆与四粒红间多态性最高,共筛选到56个多态性标记位点;秦皇岛光秧与印度窄叶间多态率最低,筛选到21个多态性标记位点。在123个多态性 SSR标记位点中,80%多态性 SSR位点片段长度为150~300 bp;18.8%多态标记的重复基序为 GA,其次为 AG、TC,分别占12.5%,12.5%;多态标记中二核苷酸、三核苷酸重复基序的重复次数在15次以上的占60%。

Through screening polymorphic SSRs and analyzing its characteristic,it is anticipated that the identi-fied set of highly informative markers should be very useful to initiate constructing SSR genetic linkage map,QTL research,and molecular assistant breeding studies in cultivated groundnut. A total of 1 523 markers designed by dif-ferent methods were used to screen for polymorphism in 6 peanut accessions with different botanical types,and the characteristics of polymorphic SSR were analyzed in the aspect of amplified fragment length,repeat motif,and repeat number. The result showed that only 43(2. 8%)pairs of markers with 123 polymorphic loci expressed polymor-phism. Among the 43 polymorphic SSRs,ARS × × showed the highest polymorphic rate(6. 8%). For the combina-tion examined,Shiyaodou and Silihong were the highest with polymorphic loci of 56,while Qinhuangdaoguangyang and Yinduzhaiye were the lowest with polymorphic loci of 21. Fragment length of 80% polymorphic loci were be-tween 150 bp and 300 bp;GA motif(18. 8%)was the most abundant,followed by AG(12. 5%)and TC(12. 5%);the repeat number of 60% of di-nucleotide and tri-nucleotide motif was over 15.
