Sevolfurane%Brain%Postoperative cognitive dysfunction%Aged%Interleukins
目的:评价七氟醚预处理对老龄大鼠术后认知功能障碍的影响及海马区炎性细胞因子含量在其中的作用。方法健康成年雄性SD大鼠138只,18个月龄,体质量400~450g,采用随机数字表法,将其分为3组(n=46)。对照组(C组):仅戊巴比妥钠麻醉,不进行手术;手术组(O组):戊巴比妥钠麻醉后行剖腹探查术,手术时间30min;七氟醚预处理组(Sev组):吸入2.4%七氟醚30min,然后吸入空气30min后处理同O组。于术前30min及术后第1、3、5、7天时,行Morris水迷宫实验,记录逃避潜伏期、平台象限停留时间和穿越原平台次数;于术前30min及术后第1、7天时,取海马组织,采用ELISA法测定海马组织TNF-α、IL-1β及IL-6含量;采用流式细胞术测定海马神经元凋亡率、胞质钙离子浓度([Ca2+]i),并于术毕即刻,采集动脉血样,行血气分析及记录体温。结果与C组比较,O组术后各时点和Sev组术后第1、3、5天时逃避潜伏期延长、平台象限停留时间缩短和穿越原平台次数减少,O组术后第1、7天和Sev组术后第1天时海马TNF-α、IL-1β及IL-6含量升高,海马神经元凋亡率、胞质钙离子浓度([Ca2+]i)升高(P<0.05);与O组比较,Sev组术后各时点逃避潜伏期缩短、平台象限停留时间延长和穿越原平台次数增多,海马TNF-α、IL-1β及IL-6含量降低,海马神经元凋亡率、胞质钙离子浓度([Ca2+]i)降低(P<0.05)。结论2.4%七氟醚预处理可减轻老龄大鼠术后认知功能障碍,其机制与降低海马区TNF-α、IL-1β及IL-6含量有关。
