目的:研究高血压患者外周血白细胞胆固醇流出相关基因表达的状况。方法选择2013年3月~2013年5月在煤炭总医院心内科门诊就诊或体检的患者。高血压患者13例,其中男性7例,女性6例,平均年龄(66.54±11.72)岁。对照组(健康体检者)11例,其中男性8例,女性3例,平均年龄(62.36±9.27)岁。采用实时荧光定量PCR方法,测定高血压患者和健康体检者外周血白细胞三磷酸腺苷结合盒转运体A1(ABCA1)、三磷酸腺苷结合盒转运体G1(ABCG1)和载脂蛋白A1(ApoAI)mRNA表达水平。结果与对照组比较,高血压组ABCA1mRNA表达降低[(2.38±0.82)vs.(0.47±0.20)],ApoAI表达也降低[(2.19±0.86)vs.(0.11±0.06)],差异有统计学意义(P均<0.05)。两组ABCG1的表达无统计学差异(P>0.05)。结论高血压患者外周血白细胞胆固醇流出基因的表达下降,这可能是高血压导致动脉粥样硬化的机制之一。

목적:연구고혈압환자외주혈백세포담고순류출상관기인표체적상황。방법선택2013년3월~2013년5월재매탄총의원심내과문진취진혹체검적환자。고혈압환자13례,기중남성7례,녀성6례,평균년령(66.54±11.72)세。대조조(건강체검자)11례,기중남성8례,녀성3례,평균년령(62.36±9.27)세。채용실시형광정량PCR방법,측정고혈압환자화건강체검자외주혈백세포삼린산선감결합합전운체A1(ABCA1)、삼린산선감결합합전운체G1(ABCG1)화재지단백A1(ApoAI)mRNA표체수평。결과여대조조비교,고혈압조ABCA1mRNA표체강저[(2.38±0.82)vs.(0.47±0.20)],ApoAI표체야강저[(2.19±0.86)vs.(0.11±0.06)],차이유통계학의의(P균<0.05)。량조ABCG1적표체무통계학차이(P>0.05)。결론고혈압환자외주혈백세포담고순류출기인적표체하강,저가능시고혈압도치동맥죽양경화적궤제지일。
Objective To study the expressions of cholesterol efflux genes in peripheral blood leukocytes in hypertensive patients.Methods The patients [hypertension group,n=13, male 7, female 6 and average age=(66.54 ±11.72)] and health cases [control group,n=11, male 8, female 3 and average age=(62.36±9.27)] were chosen from Mar. 2013 to May 2013. The mRNA expressions of apolipoprotein A1 (ApoA1), ATP binding cassette transporter A1 (ABCA1) and ATP binding cassette transporter G1 (ABCG1) in peripheral blood leukocytes were detected by using real-time fluorescence quantitative polymerase chain reaction (RT-PCR) in 2 groups.Results Compared with control group, ABCA1 mRNA expression decreased [(2.38±0.82)vs. (0.47±0.20)] and ApoAI mRNA expression also decreased [(2.19±0.86)vs. (0.11±0.06)] in hypertension group (allP<0.05). The expression of ABCG1 mRNA had no statistical difference between 2 groups (P>0.05).Conclusion The decreased expressions of cholesterol efflux genes in peripheral blood leukocytes may be one of mechanisms of hypertension resulting in atherosclerosis in hypertensive patients.
