目的:观察丹参注射液对脑梗死大鼠双侧脑室室管膜下区(SVZ)内神经干细胞(NSCs)分化作用的影响。方法成功制作大脑中动脉闭塞(MCAO)模型,随机分为丹参注射液组、生理盐水组,每组12只,每组再分为治疗7 d、14 d两个亚组。治疗前和治疗后7 d、14 d采用改良神经功能损害评分(mNSS)评定各组大鼠神经功能,另随机选取12只 SD大鼠为假手术组。以免疫荧光染色法检测 SVZ内溴脱氧尿嘧啶核苷(BrdU)/神经特异核蛋白(NeuN)、胶质纤维酸性蛋白(GFAP)/ NeuN双标阳性细胞。结果治疗7 d和14 d后,丹参注射液组 mNSS评分较生理盐水组下降,差异有统计学意义(P<0.05);各时间点丹参注射液组双侧 SVZ内 BrdU/NeuN双标阳性细胞较其他两组增高,差异有统计学意义(P<0.05)。结论丹参注射液能够促进 NSCs向神经元分化及改善神经功能,对脑梗死后神经重塑发挥了积极作用。

목적:관찰단삼주사액대뇌경사대서쌍측뇌실실관막하구(SVZ)내신경간세포(NSCs)분화작용적영향。방법성공제작대뇌중동맥폐새(MCAO)모형,수궤분위단삼주사액조、생리염수조,매조12지,매조재분위치료7 d、14 d량개아조。치료전화치료후7 d、14 d채용개량신경공능손해평분(mNSS)평정각조대서신경공능,령수궤선취12지 SD대서위가수술조。이면역형광염색법검측 SVZ내추탈양뇨밀정핵감(BrdU)/신경특이핵단백(NeuN)、효질섬유산성단백(GFAP)/ NeuN쌍표양성세포。결과치료7 d화14 d후,단삼주사액조 mNSS평분교생리염수조하강,차이유통계학의의(P<0.05);각시간점단삼주사액조쌍측 SVZ내 BrdU/NeuN쌍표양성세포교기타량조증고,차이유통계학의의(P<0.05)。결론단삼주사액능구촉진 NSCs향신경원분화급개선신경공능,대뇌경사후신경중소발휘료적겁작용。
Objective To investigate the effects of Danshen injection on differentiation of neural stem cel s (NSCs)in bilateral ventri-cle subventricular zone(SVZ)in rats with cerebral infarction.Methods The 24 successful middle cerebral artery occlusion(MCAO) adult male Sprague Dawley (SD)rat models were randomly devided into a Danshen injection group,a saline group. In each group, 12 MCAO rats were randomly al ocated into groups(6 rats in each subgroups).Neurological deficits were evaluated using the modified neurological severity score(mNSS)before treatment and on the 7 th,14 th day after treatment.Another randomly selected 12 normal rats served as sham operation group. BrdU/NeuN,NeuN/GFAP double positive cel s were detected by immunofluorescence tech-niques.Results After 7 d,14 d treatment,Danshen injection group rats’mNSS was decreased compared with saline group,the difference was statistical y significant(P<0.05);each time,the number of BrdU+ /NeuN+ cel s in Danshen injection group were in-creased than the other two groups,the difference was statistical y significant(P<0.05).Conclusion Danshen injection can promote differentiation of NSCs into neurons and improve neurological function after cerebral infarction,play a positive role in neural remodeling.
