Myocardial remodeling associated factors%Sympathetic nervous system%Yiqiwenyanghuoxue decoction
目的:研究益气温阳活血方对慢性心力衰竭大鼠心室重塑相关因子的影响。方法通过异丙肾上腺素(ISO)诱导慢性心力衰竭动物模型。选择健康Wistar大鼠40只,随机分为正常组、模型组、卡维地洛组(西药组)、益气温阳活血方组(中药组);心脏超声检测心脏结构及功能,以酶联免疫吸附法检测去甲肾上腺素(N E )、血管紧张素Ⅱ(AngⅡ)、内皮素‐1(ET‐1)、一氧化氮(NO)、金属基质蛋白酶‐2(MMP‐2)、金属基质蛋白酶‐9(MMP‐9)、金属基质蛋白酶抑制物‐1(TIMP‐1)、金属基质蛋白酶抑制物‐2(TIMP‐2)含量。结果模型大鼠左室后壁厚度(LVPW)、室间隔厚度(IVS)明显升高,射血分数(EF%值)则明显降低;NE、AngⅡ、ET‐1、MMP‐2、MMP‐9含量明显升高,而NO、TIMP‐1、TIMP‐2含量则明显降低,与正常组比较差异有统计学意义(P<0.05);益气温阳活血方可降低慢性心力衰竭大鼠NE、AngⅡ、ET‐1、MMP‐2、MMP‐9含量及升高NO、TIMP‐1、TIMP‐2含量,提高EF%值、降低IVS及LVPW厚度,与模型组比较差异有统计学意义(P<0.05),而与正常组及卡维地洛组比较差异无统计学意义(P>0.05)。结论益气温阳活血方可抑制心力衰竭大鼠相关心室重塑因子的水平,防止心室重塑。

목적:연구익기온양활혈방대만성심력쇠갈대서심실중소상관인자적영향。방법통과이병신상선소(ISO)유도만성심력쇠갈동물모형。선택건강Wistar대서40지,수궤분위정상조、모형조、잡유지락조(서약조)、익기온양활혈방조(중약조);심장초성검측심장결구급공능,이매련면역흡부법검측거갑신상선소(N E )、혈관긴장소Ⅱ(AngⅡ)、내피소‐1(ET‐1)、일양화담(NO)、금속기질단백매‐2(MMP‐2)、금속기질단백매‐9(MMP‐9)、금속기질단백매억제물‐1(TIMP‐1)、금속기질단백매억제물‐2(TIMP‐2)함량。결과모형대서좌실후벽후도(LVPW)、실간격후도(IVS)명현승고,사혈분수(EF%치)칙명현강저;NE、AngⅡ、ET‐1、MMP‐2、MMP‐9함량명현승고,이NO、TIMP‐1、TIMP‐2함량칙명현강저,여정상조비교차이유통계학의의(P<0.05);익기온양활혈방가강저만성심력쇠갈대서NE、AngⅡ、ET‐1、MMP‐2、MMP‐9함량급승고NO、TIMP‐1、TIMP‐2함량,제고EF%치、강저IVS급LVPW후도,여모형조비교차이유통계학의의(P<0.05),이여정상조급잡유지락조비교차이무통계학의의(P>0.05)。결론익기온양활혈방가억제심력쇠갈대서상관심실중소인자적수평,방지심실중소。
Objective To study effects of Yiqiwenyanghuoxue decoction on myocardial remodeling associated fac‐tors of rat with chronic heart failure .Method The model rats with chronic heart failure were injected with ISO .40 healthy Wistarrats were randomly separated into 4 groups:normal group ,ISO‐injected group ,ISO‐injected with Carvedilol group and ISO‐injected with Yiqiwnyanghoxue group ,then cardiac structure and function were ?tested by echocardiography ,AngⅡ ,ET‐1 ,NO ,MMP‐2 ,MMP‐9 ,TIMP‐1 and TIMP‐2 were measured by ELISA .Re‐sult Compared with the normal group ,those were significantly higher at LVPW ,IVS NE ,NE ,Ang Ⅱ ,ET‐1 , MMP‐2 and MMP‐9 in ISO‐injected group ,but those were significantly lower at EF% ,NO ,TIMP‐1 and TIMP‐2 (all P<0 .05) ,and these change can be attenuated by decoction of yiqiwenyanghuoxue ,compared with the normal group and ISO‐injected with Carvediol group ,there were no significant differences in ISO‐injected with Yiqiwn‐yanghoxue group (all P> 0 .05) .Conclusion Yiqiwenyanghuoxue decoction may reverse the myocardial remodeling through regulating myocardial remodeling associated factors .
