目的:皮质注射氯化亚铁(FeCl2)致外伤性癫痫(PTE)动物模型,探讨 FeCl2致痫的可能机制。方法:成年 SD 大鼠24只随机分为正常组和模型组,各12只。通过注射 FeCl2制作 PTE 模型,观察各组癫痫发作的行为、脑电图及病理表现,RT-PCR 法检测海马及额叶 c-fos,c-jun 及 c-myc mRNA 转录水平。结果:模型组在行为学及脑电图表现均接近临床癫痫发作;模型组中 c-fos、c-jun mRNA 转录水平与正常组差异无统计学意义(P>0.05);模型组中 c-myc mRNA 转录水平显著高于正常组(P<0.01)。结论:FeCl2皮质注射制作 PTE 的机制可能与 c-myc mRNA 转录水平的升高有关。

목적:피질주사록화아철(FeCl2)치외상성전간(PTE)동물모형,탐토 FeCl2치간적가능궤제。방법:성년 SD 대서24지수궤분위정상조화모형조,각12지。통과주사 FeCl2제작 PTE 모형,관찰각조전간발작적행위、뇌전도급병리표현,RT-PCR 법검측해마급액협 c-fos,c-jun 급 c-myc mRNA 전록수평。결과:모형조재행위학급뇌전도표현균접근림상전간발작;모형조중 c-fos、c-jun mRNA 전록수평여정상조차이무통계학의의(P>0.05);모형조중 c-myc mRNA 전록수평현저고우정상조(P<0.01)。결론:FeCl2피질주사제작 PTE 적궤제가능여 c-myc mRNA 전록수평적승고유관。
Objective: To establish FeCl2-induced post traumatic epilepsy (PTE) model, and to explore the pos-sible mechanisms. Methods: Twenty-four healthy adult SD rats were randomly divided into control group (n=12) and model group (n=12). The PTE animal model was established by injecting FeCl2 into the cerebral cortex. Changes in behaviors, electroencephalograms (EEG) and histology of the hippocampus and frontal lobes were stud-ied. The expression levels of c-fos, c-jun and c-myc mRNA were tested. Result: The behaviors and EEG of the rats in the model group were similar to the clinical feature of human epileptic seizures. There was no statistical difference in the expression levels of c-fos and c-jun mRNA between the normal control group and model group (P>0.05). As compared with the control group, the expression level of c-myc mRNA in the model group was increased significantly (P<0.01). Conclusion: The FeCl2-induced PTE model may be meditaed by the increase of the expression level of c-myc mRNA.
