目的:探讨骨髓间充质干细胞(MSC)移植治疗对大鼠脑缺血后神经功能及脑组织中 IL-10、TNF-α表达的影响。方法:原代分离培养 MSC。27只大鼠随机分为3组,各9只:假手术组,仅剪开皮肤;模型组,制作局灶性大脑中动脉闭塞(MCAO)模型,术后24 h 尾静脉注射 PBS;MSC 组,制作 MCAO 模型,术后24 h 尾静脉注射移植 MSC。术后第3天行改良大鼠神经功能缺损评分(mNSS)评定大鼠神经功能;行为学测试后取梗死灶周围缺血半暗带脑组织,western blot 法检测肿瘤坏死因子(TNF)-α、白细胞介素(IL)-10蛋白表达水平。结果:与模型组比较,MSC 组 mNSS 评分升高(P<0.05),脑组织蛋白 IL-10表达水平上调(P<0.05),TNF-α表达水平下调(P<0.05)。结论:尾静脉移植 MSC 治疗可改善局灶性脑缺血大鼠神经功能,可能与移植 MSC后脑组织中 TNF-α和 IL-10水平有关。
目的:探討骨髓間充質榦細胞(MSC)移植治療對大鼠腦缺血後神經功能及腦組織中 IL-10、TNF-α錶達的影響。方法:原代分離培養 MSC。27隻大鼠隨機分為3組,各9隻:假手術組,僅剪開皮膚;模型組,製作跼竈性大腦中動脈閉塞(MCAO)模型,術後24 h 尾靜脈註射 PBS;MSC 組,製作 MCAO 模型,術後24 h 尾靜脈註射移植 MSC。術後第3天行改良大鼠神經功能缺損評分(mNSS)評定大鼠神經功能;行為學測試後取梗死竈週圍缺血半暗帶腦組織,western blot 法檢測腫瘤壞死因子(TNF)-α、白細胞介素(IL)-10蛋白錶達水平。結果:與模型組比較,MSC 組 mNSS 評分升高(P<0.05),腦組織蛋白 IL-10錶達水平上調(P<0.05),TNF-α錶達水平下調(P<0.05)。結論:尾靜脈移植 MSC 治療可改善跼竈性腦缺血大鼠神經功能,可能與移植 MSC後腦組織中 TNF-α和 IL-10水平有關。
목적:탐토골수간충질간세포(MSC)이식치료대대서뇌결혈후신경공능급뇌조직중 IL-10、TNF-α표체적영향。방법:원대분리배양 MSC。27지대서수궤분위3조,각9지:가수술조,부전개피부;모형조,제작국조성대뇌중동맥폐새(MCAO)모형,술후24 h 미정맥주사 PBS;MSC 조,제작 MCAO 모형,술후24 h 미정맥주사이식 MSC。술후제3천행개량대서신경공능결손평분(mNSS)평정대서신경공능;행위학측시후취경사조주위결혈반암대뇌조직,western blot 법검측종류배사인자(TNF)-α、백세포개소(IL)-10단백표체수평。결과:여모형조비교,MSC 조 mNSS 평분승고(P<0.05),뇌조직단백 IL-10표체수평상조(P<0.05),TNF-α표체수평하조(P<0.05)。결론:미정맥이식 MSC 치료가개선국조성뇌결혈대서신경공능,가능여이식 MSC후뇌조직중 TNF-α화 IL-10수평유관。
Objective: To investigate the effects of transplantation of bone marrow mesenchymal stem cells (MSCs) on the neurological function and the expression of tumor necrosis factor-alpha (TNF-α) and interleukin-10 (IL-10) in rats after cerebral ischemia. Methods: Twenty-seven SD rats were randomly divided into 3 groups, sham group, ischemia group and MSC group (n=9 respectively). The middle cerebral artery occlusion (MCAO) model was induced with filament occlusion method. Rats in the ischemia group were transplanted with primarily cultured MSCs 24 h after MCAO. Neural function was evaluated and the expression of TNF-α and IL-10 were assayed at day 3 after operation. Results: When compared with those in the ischemia group, the neural function was signifi-cantly improved (P<0.05), the level of TNF-α protein was significantly decreased (P<0.05) and the level of IL-10 protein was significantly increased (P<0.05) of those in MSC group. Conclusion: Transplantation of MSCs could promote the nerve function recovery after cerebral ischemia.
