Medicinal Fritillaria from Xinjiang%total alkaloids%quantitative determination
目的::探讨新疆贝母属药用贝母中总生物碱含量测定的最佳方法,并比较其总生物碱含量差异。方法:用氯仿∶甲醇(4∶1)混合溶剂提取总生物碱;采用酸性染料比色法测定新疆贝母属药用贝母中总生物碱含量。结果:酸性染料选择溴麝香草酚蓝比色的显色条件为 pH 5.0缓冲液4.0 mL;氯仿萃取1次,每次2.6 mL;无水硫酸钠脱水用量0.25 g。西贝素在12.24~73.44μg 范围内呈良好的线性关系;平均回收率101.53%,RSD 为2.67%(n =5)。所测生物碱的含量在新疆贝母属药用贝母种间存在一定差异,除滩贝母和小花贝母总生物碱含量相对较低外,其余6种贝母总生物碱含量在0.589%~0.869%之间,比以往文献测定的总生物碱含量均较高。结论:所建立的方法简便、精确,重复性好,可用于新疆贝母属药用贝母总生物碱含量的测定,其总生物碱含量为新疆贝母属药用贝母资源进一步开发利用提供科学依据。

Objective: To investigate the optimum method of determination of total alkaloids in medicinal Fritillaria from the genus Xinjiang Fritillaria, and a difference of total alkaloids contents in different spe-cies medicinal Fritillaria from the genus Xinjiang Fritillaria were compared. Methods: The contents of to-tal alkaloid in medicinal Fritillaria from the genus Xinjiang Fritillaria were determined by acid dye colori-metry. Results: Bromothymol blue was selected as acid dye colorimetry, and acid dye color conditions:pH 5. 0 buffer 4. 0 mL. Chloroform extraction with 2. 6 mL, and anhydrous sodium sulfate 0. 25 g. Sipei-mine in 12. 24 ~ 73. 44 μg range good linear relationship,average recovery was 101. 53% ,RSD was 2. 67% (n = 5). There existed a little differences in total alkaloids contents in different species medicinal Fritillaria from the genus Xinjiang Fritillaria. Total alkaloids contents in 6 species medicinal Fritillaria from the genus Xinjiang Fritillaria were from 0. 589 to 0. 869 percent, while Total alkaloids contents in F. karelinii (Fisch. ) Baker. and F. meleagroides were relatively lower. Generally speaking, they all was relatively higher in total alkaloids contents between this study and previous literature. Conclusion:The method is simple,accurate, reproducible and can be used to determine total alkaloids in medicinal Fritillaria from the genus Xinjiang Fritillaria. This provided a scientific basis for exploitation and utiliza-tion of medicinal Fritillaria from the genus Xinjiang Fritillaria in the future with high content of alkaloids.
