目的:通过观察补体 C3在大鼠肝脏缺血再灌注模型中的变化,探讨补体 C3在大鼠肝脏缺血再灌注中的作用。方法健康雄性SD大鼠60只,180~200 g;随机分成假手术组(S组),全肝缺血再灌注1h组(I/R1h组),全肝缺血再灌注3h组(I/R3h组),全肝缺血再灌注6h组(I/R 6 h组)和全肝缺血再灌注24 h组(I/R 24 h组),共5组,每组12只。建立全肝缺血再灌注模型,于再灌注时1、3、6、24 h选取肝脏标本行HE染色,在显微镜下观察肝脏的病理学改变,荧光实时定量PCR法检测补体C3 mRNA表达,Western blot法检测补体C3的表达。结果(1)S组可以观察到肝脏组织的结构基本正常,I/R(1 h,3 h,6 h,24 h)4组均有不同程度肝组织的损伤,并且损伤程度随着缺血再灌注的时间延长而逐渐加重。(2)与S组比较,I/R 1 h组的补体C3 mRNA的表达差异无统计学意义(P>0.05);随着时间延长,补体C3 mRNA的表达逐渐升高,于24 h时达到最高峰,与其他4组比较具有明显的统计学意义(P<0.05);补体C3的蛋白表达也随着时间的延长逐渐升高,I/R 1 h组的蛋白表达与S组比较无明显上调(P>0.05),补体C3蛋白含量在I/R 24 h组中表达达到高峰,与其他I/R组相差异有统计学意义(P<0.05)。结论补体C3的活化参与了大鼠肝脏缺血再灌注损伤。

목적:통과관찰보체 C3재대서간장결혈재관주모형중적변화,탐토보체 C3재대서간장결혈재관주중적작용。방법건강웅성SD대서60지,180~200 g;수궤분성가수술조(S조),전간결혈재관주1h조(I/R1h조),전간결혈재관주3h조(I/R3h조),전간결혈재관주6h조(I/R 6 h조)화전간결혈재관주24 h조(I/R 24 h조),공5조,매조12지。건립전간결혈재관주모형,우재관주시1、3、6、24 h선취간장표본행HE염색,재현미경하관찰간장적병이학개변,형광실시정량PCR법검측보체C3 mRNA표체,Western blot법검측보체C3적표체。결과(1)S조가이관찰도간장조직적결구기본정상,I/R(1 h,3 h,6 h,24 h)4조균유불동정도간조직적손상,병차손상정도수착결혈재관주적시간연장이축점가중。(2)여S조비교,I/R 1 h조적보체C3 mRNA적표체차이무통계학의의(P>0.05);수착시간연장,보체C3 mRNA적표체축점승고,우24 h시체도최고봉,여기타4조비교구유명현적통계학의의(P<0.05);보체C3적단백표체야수착시간적연장축점승고,I/R 1 h조적단백표체여S조비교무명현상조(P>0.05),보체C3단백함량재I/R 24 h조중표체체도고봉,여기타I/R조상차이유통계학의의(P<0.05)。결론보체C3적활화삼여료대서간장결혈재관주손상。
Objective To research and analyze the role of the complement C3 by observing the change of complement C3 in the model of hepatic ischemia reperfusion. Methods Sixty healthy and male SD rats, each weighing180-200 g, were randomly assigned to sham operation group (group S), hepatic I/R 1 hour (I/R 1 h group), hepatic I/R 3 hours (I/R 3 h group), hepatic I/R 6 hours (I/R 6 h group), hepatic I/R 24 hours (I/R 24 h group). There were 5 groups, 12 rats in each group, which built the model of hepatic ischemia reperfusion. Hepatic specimens were obtained at 1, 3, 6, and 24 h of reperfusion and stained with haematoxylin and eosin for examination of histological changes of the liver, and for expression of complement C3 mRNA by using real-time PCR and for expression of complement C3 by using Western blot. Results The structure of liver tissue was basically normal from the observation of group S, while the structures of liver tissue from I/R(1 h, 3 h, 6 h, 24 h) were injured in varying degrees and the degrees of injury were aggravated with the prolonging time of reperfusion. Compared with group S, the expression of complement C3 mRNA had no statistics meanings (P>0.05);with the prolonging of time, the expression of complement C3 mRNA was higher and higher and reached the peak at 24 hours, which had the obvious statistics meanings compared with the other four groups (P<0.05); the expression of complement C3’protein was higher and higher with the prolonging of time, and compared with group S, the expression of protein from I/R 24 hour was not clearly increased (P>0.05). the content of the expression of complement C3 protein reached the peak in the group I/R 24 h, and compared with other I/R group, it had the significant difference and statistics meanings (P<0.05). Conclusion Activation of complement C3 is involved in hepatic I/R injury rats.
