目的:通过孕期中给予孕鼠不同的葡萄糖摄入量,研究其胎盘中胰岛素样生长因子(IGF)表达情况,同时观察胎鼠结局。方法选取成功交配的雌性KM小鼠48只,随机分为3组:对照组(8只)、实验A组(20只)和实验B组(20只)。孕期中三组均给予正常喂养,A组每天另给予5%葡萄糖溶液1 ml灌胃,B组另给予10%葡萄糖溶液1 ml灌胃。所有孕鼠均在第19天时进行剖宫产,使用Western blot和RT-PCR方法对胎盘组织的IGF-1、IGF-2及IGF-1R进行检测,同时对胎鼠数量和畸形、死胎、存活胎情况进行观察,并对存活胎胎鼠体重和血糖水平进行检测。结果(1)Western blot和RT-PCR检测结果显示,实验A组和实验B组孕鼠胎盘组织中IGF-1、IGF-2及IGF-1R的表达水平均高于对照组,其中实验B组中的表达水平最高(P<0.05);(2)实验A组和实验B组的胎鼠体重和血糖水平均高于对照组,其中实验B组水平最高(P<0.05);(3)三组中胎鼠发育均较正常,胎鼠数量和畸形、死胎情况未发现有明显增多(P>0.05)。结论孕鼠葡萄糖摄入量增多会诱导胎盘组织IGF-1、IGF-2及IGF-1R表达水平增加,促进胎鼠对葡萄糖的吸收利用及胎儿体重的增加。

목적:통과잉기중급여잉서불동적포도당섭입량,연구기태반중이도소양생장인자(IGF)표체정황,동시관찰태서결국。방법선취성공교배적자성KM소서48지,수궤분위3조:대조조(8지)、실험A조(20지)화실험B조(20지)。잉기중삼조균급여정상위양,A조매천령급여5%포도당용액1 ml관위,B조령급여10%포도당용액1 ml관위。소유잉서균재제19천시진행부궁산,사용Western blot화RT-PCR방법대태반조직적IGF-1、IGF-2급IGF-1R진행검측,동시대태서수량화기형、사태、존활태정황진행관찰,병대존활태태서체중화혈당수평진행검측。결과(1)Western blot화RT-PCR검측결과현시,실험A조화실험B조잉서태반조직중IGF-1、IGF-2급IGF-1R적표체수평균고우대조조,기중실험B조중적표체수평최고(P<0.05);(2)실험A조화실험B조적태서체중화혈당수평균고우대조조,기중실험B조수평최고(P<0.05);(3)삼조중태서발육균교정상,태서수량화기형、사태정황미발현유명현증다(P>0.05)。결론잉서포도당섭입량증다회유도태반조직IGF-1、IGF-2급IGF-1R표체수평증가,촉진태서대포도당적흡수이용급태인체중적증가。
Objective To investigate the placental IGFs expressions and fetal rats results when pregnant rats intake difference levels of glucose in pregnancy. Methods There were 48 female KM rats which had copulated successfully were selected, they were randomly divided into three groups:the control group (n=8), the experimental group A (n=20) and the experimental group B (n=20). During pregnancy, the three groups rats were given normal feeding, the experimental group A was given 5% glucose solution intragastric administration every day, the experimental group B was given 10% glucose solution intragastric administration every day. All the pregnant rats were given cesarean at the 19th days, Western blot and RT-PCR method were used to detect IGF-1, IGF-2 and IGF-1R of placental tissue expression levels, while the number of fetal, malformation, stillbirth and survival of fetal were observed, survival of fetal body weight and blood glucose level were detected. Results (1) Western blot and RT-PCR detecting results showed that IGF-1, IGF-2 and IGF-1R expression levels in the experimental group A and B were higher than the control group, the experimental group B was the highest (P<0.05); (2) Fetal rats body weight and blood glucose level in the experimental group A and B were higher than the control group, the experimental group B was the highest (P<0.05);(3) Fetal rats development were normal in three groups, the number of fetal rats, malformation and stillbirth were not reach difference (P>0.05). Conclusion The high level glucose intake during pregnancy may result in higher placental tissue IGF-1 and IGF-2 and IGF-1R expression levels, which can promote fetal rats glucose utilization and weight gain.
