目的:观察脂氧素 A4预处理对链尿佐菌素诱导的糖尿病大鼠心肌缺血再灌注时 Caspase -3、Bcl -2和 Bax 表达的影响。方法雄性 SD 大鼠60只,随机分为 CS、CI/R、CLX、DS、DI/R、DLX 组共6组(n =10)。 CS、CI/R、CLX组采用腹腔注射链尿佐菌素55 mg/kg 制备糖尿病模型。 CI/R、DI/R组建立大鼠心肌缺血再灌注模型,结扎冠脉左前降支30 min,CS 组和 DS 组仅穿线不结扎,CI/R组和 DI/R组缺血前5 min 经股静脉注射2 mL/kg 生理盐水,CLX组和 DLX组缺血前5 min 经股静脉注射脂氧素 A42.5μg/kg。再灌注4 h 时处死大鼠,取心肌组织,采用 HE 染色法光镜下观察缺血心肌组织形态学改变,通过 RT -PCR 和 Western blot 法检测缺血心肌 Caspase -3、Bcl -2和 Bax的表达水平,并计算 Bcl -2与 Bax 表达的比值(Bcl -2/Bax)。结果CLX组和 DLX组心肌组织病理学损伤较 CI/R组和 DI/R组减轻,Caspase -3和 Bax 的表达量减少,Bcl -2的表达量增加,Bcl -2/Bax 比值升高(P <0.05)。结论脂氧素 A4预处理能够下调糖尿病大鼠心肌缺血再灌注时 Caspase -3和 Bax 的表达,上调 Bcl -2的表达,使 Bcl -2/Bax 比值升高。

목적:관찰지양소 A4예처리대련뇨좌균소유도적당뇨병대서심기결혈재관주시 Caspase -3、Bcl -2화 Bax 표체적영향。방법웅성 SD 대서60지,수궤분위 CS、CI/R、CLX、DS、DI/R、DLX 조공6조(n =10)。 CS、CI/R、CLX조채용복강주사련뇨좌균소55 mg/kg 제비당뇨병모형。 CI/R、DI/R조건립대서심기결혈재관주모형,결찰관맥좌전강지30 min,CS 조화 DS 조부천선불결찰,CI/R조화 DI/R조결혈전5 min 경고정맥주사2 mL/kg 생리염수,CLX조화 DLX조결혈전5 min 경고정맥주사지양소 A42.5μg/kg。재관주4 h 시처사대서,취심기조직,채용 HE 염색법광경하관찰결혈심기조직형태학개변,통과 RT -PCR 화 Western blot 법검측결혈심기 Caspase -3、Bcl -2화 Bax적표체수평,병계산 Bcl -2여 Bax 표체적비치(Bcl -2/Bax)。결과CLX조화 DLX조심기조직병이학손상교 CI/R조화 DI/R조감경,Caspase -3화 Bax 적표체량감소,Bcl -2적표체량증가,Bcl -2/Bax 비치승고(P <0.05)。결론지양소 A4예처리능구하조당뇨병대서심기결혈재관주시 Caspase -3화 Bax 적표체,상조 Bcl -2적표체,사 Bcl -2/Bax 비치승고。
Objective To investigate the effects of lipoxin A4 pre -treatment on expression of Caspase -3, Bcl -2 and Bax following myocardial ischemia -reperfusion injury (MIRI) in diabetic mellitus rats.Methods Sixty male Spra-gue -Dawley rats were randomly divided into 6 groups (n =10 each), normal rats sham operation group (CS ), normal rats myocardial I /R group (CI/R ), normal rats lipoxin A4 group (CLX ), diabetic rats sham operation group (DS ), diabet-ic rats myocardial I /R group (DI/R ) and diabetic rats lipoxin A4 group (DLX ).Diabetic mellitus was induced by intraper -itoneal STZ injection.MIRI was induced by 30 min occlusion of left anterior descending branch of coronary artery followed with 4 hours reperfusion.Rats were then sacrificed and myocardial specimens were isolated .The myocardial pathological pattern was detected by HE staining.The expression of Caspase -3, Bcl -2 and Bax was assessed by semi -quantitative reverse transcription polymerase chain reaction (RT -PCR) and Western blot methods, as well as the Bcl -2/Bax ratio was also calculated.Results The myocardial damages in Group C LX and Group DLX were significantly reduced.Significant down -regulation of Caspase -3 and Bax, and increase of Bcl -2 and Bcl -2/Bax ratio were revealed in Group C LX and Group DLX while compared with Group C I/R and Group D I/R , respectively.Conclusion Lipoxin A4 pretreament down -regulates the expression of Caspase -3 and Bax, up -regulates Bcl -2 expression, and increases the Bcl -2 /Bax ratio.
