目的:研究没药甾酮对胆管癌RBE细胞株移植瘤生长和miRNA表达的影响。方法雌性SD大鼠建立胆管癌RBE细胞株移植瘤模型,随机分为对照组和没药甾酮高、低剂量实验组;高、低剂量实验组大鼠分别予40、20 mg· kg-1· d-1没药甾酮灌胃,对照组大鼠予生理盐水灌胃。每5天测量裸鼠质量和肿瘤体积,处理30 d后取材,提取肿瘤组织总RNA并分离miR-NA。采用微阵列杂交,通过芯片扫描和数据分析获得miRNAs表达谱。结果高、低剂量实验组大鼠移植瘤体积分别在处理20 d和25 d后显著低于对照组(P<0.01);处理30 d 后高、低剂量组移植瘤质量均显著低于对照组(P<0.01)。芯片结果显示,高剂量实验组瘤组织miRNAs表达谱与对照组相比有显著差异。结论没药甾酮可以显著抑制胆管癌RBE细胞株移植瘤生长并影响其miRNA的表达。
目的:研究沒藥甾酮對膽管癌RBE細胞株移植瘤生長和miRNA錶達的影響。方法雌性SD大鼠建立膽管癌RBE細胞株移植瘤模型,隨機分為對照組和沒藥甾酮高、低劑量實驗組;高、低劑量實驗組大鼠分彆予40、20 mg· kg-1· d-1沒藥甾酮灌胃,對照組大鼠予生理鹽水灌胃。每5天測量裸鼠質量和腫瘤體積,處理30 d後取材,提取腫瘤組織總RNA併分離miR-NA。採用微陣列雜交,通過芯片掃描和數據分析穫得miRNAs錶達譜。結果高、低劑量實驗組大鼠移植瘤體積分彆在處理20 d和25 d後顯著低于對照組(P<0.01);處理30 d 後高、低劑量組移植瘤質量均顯著低于對照組(P<0.01)。芯片結果顯示,高劑量實驗組瘤組織miRNAs錶達譜與對照組相比有顯著差異。結論沒藥甾酮可以顯著抑製膽管癌RBE細胞株移植瘤生長併影響其miRNA的錶達。
목적:연구몰약치동대담관암RBE세포주이식류생장화miRNA표체적영향。방법자성SD대서건립담관암RBE세포주이식류모형,수궤분위대조조화몰약치동고、저제량실험조;고、저제량실험조대서분별여40、20 mg· kg-1· d-1몰약치동관위,대조조대서여생리염수관위。매5천측량라서질량화종류체적,처리30 d후취재,제취종류조직총RNA병분리miR-NA。채용미진렬잡교,통과심편소묘화수거분석획득miRNAs표체보。결과고、저제량실험조대서이식류체적분별재처리20 d화25 d후현저저우대조조(P<0.01);처리30 d 후고、저제량조이식류질량균현저저우대조조(P<0.01)。심편결과현시,고제량실험조류조직miRNAs표체보여대조조상비유현저차이。결론몰약치동가이현저억제담관암RBE세포주이식류생장병영향기miRNA적표체。
Objective To determine the effects of guggulsterone on the growth and microRNAs expression profile in transplanted tumor of human cholangiocarcinoma RBE cells.Methods Female SD nude mice were injected with RBE cells to establish transplanted tumor models.The mice were equally randomized into 3 groups including control group,high dose and low dose treatment groups.Oral admin-istration of guggulsterone [0, 40 and 20 mg· kg-1 · d-1 respectively] were given to the corresponding mice in 72 h after transplanta-tion.Tumor volumes and weights were measured every 5 d.Mice were sacrificed after 30 days’ treatment.Total RNA was extracted and miRNAs were isolated and hybridized with miRNAs microarrays.Results The growth of transplanted tumors was significantly sup-pressed in treatment groups.Tumors volume were significantly smaller in high dose group and low dose group than in the control after 20 days’(P<0.01) and 25 days’treatment (P <0.01).The microarray assay showed significantly different miRNAs expression in tumors of high dose group compared with the control group Conclusion Guggulsterone significantly suppresses the growth of transplan-ted tumor of human cholangiocarcinoma RBE cells and affect microRNAs expression profile.
