目的:观察漆树酸对人前列腺癌细胞 LNCaP 细胞周期的影响并分析其机制。方法人前列腺癌细胞 LNCaP 经漆树酸处理后,MTS 法观察不同时间、不同浓度的漆树酸对细胞增殖率的影响,流式细胞术分析细胞周期变化,Western blot 方法观察漆树酸作用后细胞周期相关因子 CDK4、CDK6、cyclin D1和 P27蛋白水平的表达变化。结果(1)漆树酸时间和浓度依赖性地抑制细胞增殖;(2)经漆树酸处理后的细胞出现生长抑制,细胞主要阻滞在 G0/G1期;(3)漆树酸作用细胞后,CDK4、CDK6、cyclin D1蛋白表达水平低于溶剂对照组,而 P27蛋白表达水平高于溶剂对照组。结论漆树酸可抑制 LNCaP 细胞增殖,其机制可能是与细胞周期相关蛋白 P27的上调, CDK4、CDK6、cyclin D1表达下调有关,导致细胞阻滞在 G0/G1期。

목적:관찰칠수산대인전렬선암세포 LNCaP 세포주기적영향병분석기궤제。방법인전렬선암세포 LNCaP 경칠수산처리후,MTS 법관찰불동시간、불동농도적칠수산대세포증식솔적영향,류식세포술분석세포주기변화,Western blot 방법관찰칠수산작용후세포주기상관인자 CDK4、CDK6、cyclin D1화 P27단백수평적표체변화。결과(1)칠수산시간화농도의뢰성지억제세포증식;(2)경칠수산처리후적세포출현생장억제,세포주요조체재 G0/G1기;(3)칠수산작용세포후,CDK4、CDK6、cyclin D1단백표체수평저우용제대조조,이 P27단백표체수평고우용제대조조。결론칠수산가억제 LNCaP 세포증식,기궤제가능시여세포주기상관단백 P27적상조, CDK4、CDK6、cyclin D1표체하조유관,도치세포조체재 G0/G1기。
Objective To observe the effects and mechanism on cell cycle of anacardic acid in human prostate cancer LNCaP cells.Methods MTS was used to detect cell proliferation rate of LNCaP cells treated with different con -centraions of anacardic acid at different time spots .The cell cycle analysis was determined by flow cytometry , and the changes of cell cycle related CDK4, CDK6, cyclin D1 and P27 were examined by Western blot.Results (1) MTS: the cell proliferation was significantly inhibited by anacardic acid in time -and dose -dependent manners; (2) Flow cytome-try: after treated by anacardic acid, cells growth were inhibited and mainly blocked at G 0 /G1 phase.(3) Western blot:after treated by anacardic acid , CDK4, CDK6 and cyclin D1 were down -regulated while P27 up -regulated.Conclusion Anacardic acid can inhibit LNCaP cell proliferation , which is mediated by the up -regulation of cell cycle -related pro-tein P27, the down -regulation of CDK4, CDK6 and cyclin D1, and the G0 /G1 phase arrest.
