目的:探讨 miR-584对人胶质母细胞瘤 U251细胞侵袭和迁移能力的影响。方法将化学合成的 miR-584-3p 在脂质体 Lipofectamine 2000的介导下,瞬时转染人胶质母细胞瘤 U251,细胞分为空白对照组(无转染,其他条件相同)、mimics NC 组(转染 miR-584-3p mimics 阴性对照序列)、miR-584-3p mimics 组(转染 miR-584-3p mim-ics)、inhibitor NC 组(转染 miR-584-3p inhibitor 阴性对照序列)和 miR-584-3p inhibitor 组(转染 miR-584-3p inhibi-tor)。Real-time PCR 法检测细胞中 miR-584-3p 的表达水平;CCK-8法检测细胞增殖能力;划痕实验和 Transwell小室实验检测细胞的迁移和侵袭能力。结果与空白对照组和 mimics NC 组相比,miR-584-3p mimics 组的 miR-584-3p 表达上调约100倍(P <0.05),细胞增殖能力无明显差异(P >0.05),细胞侵袭和迁移能力明显下降(P <0.05);与空白对照组和 inhibitor NC 组相比,miR-584-3p inhibitor 组的 miR-584-3p 表达下调至空白对照组的5%(P <0.05),细胞增殖能力无明显差异(P >0.05),细胞侵袭和迁移能力显著增强(P <0.05)。结论miR-584-3p mimics转染抑制了 U251细胞的侵袭和迁移能力;miR-584-3p inhibitor 转染能增强 U251细胞的侵袭和迁移能力。

목적:탐토 miR-584대인효질모세포류 U251세포침습화천이능력적영향。방법장화학합성적 miR-584-3p 재지질체 Lipofectamine 2000적개도하,순시전염인효질모세포류 U251,세포분위공백대조조(무전염,기타조건상동)、mimics NC 조(전염 miR-584-3p mimics 음성대조서렬)、miR-584-3p mimics 조(전염 miR-584-3p mim-ics)、inhibitor NC 조(전염 miR-584-3p inhibitor 음성대조서렬)화 miR-584-3p inhibitor 조(전염 miR-584-3p inhibi-tor)。Real-time PCR 법검측세포중 miR-584-3p 적표체수평;CCK-8법검측세포증식능력;화흔실험화 Transwell소실실험검측세포적천이화침습능력。결과여공백대조조화 mimics NC 조상비,miR-584-3p mimics 조적 miR-584-3p 표체상조약100배(P <0.05),세포증식능력무명현차이(P >0.05),세포침습화천이능력명현하강(P <0.05);여공백대조조화 inhibitor NC 조상비,miR-584-3p inhibitor 조적 miR-584-3p 표체하조지공백대조조적5%(P <0.05),세포증식능력무명현차이(P >0.05),세포침습화천이능력현저증강(P <0.05)。결론miR-584-3p mimics전염억제료 U251세포적침습화천이능력;miR-584-3p inhibitor 전염능증강 U251세포적침습화천이능력。
Objective To explore the effect of miR-584 expression on the invasion and migration of glioblastoma U251 cells in vitro.Methods Chemosynthetic miR-584-3p was transfected into glioblastoma cells under the induction of Lipofectamine 2000.The cells were divided into control group(no transfection),mimics NC group(transfected with negative control sequence of miR-584-3p mimics),miR-584-3p mimics group(transfected with miR-584-3p mimics), inhibitor NC group(transfected with negative control sequence of miR-584-3p inhibitor),miR-584-3p inhibitor group (transfected with miR-584-3p inhibitor).The expression of miR-584-3p was detected by Real-time PCR.Cell prolifera-tion was detected by CCK-8 proliferation assay.The invasive and migratory effects of glioblastoma cell were measured by Wound healing and Transwell assays.Results Compared with mimics NC group and control group,the expression of miR-584-3p in miR-584-3p mimics group was upregulated by 100 times(P <0.05 ).Compared with mimics NC group and control group,the cell proliferation ability of miR-584-3p mimics group showed no significant change(P >0.05),but the cellular migratory and invasive activity of miR-584-3p mimics group were down-regulated significantly (P <0.05).Compared with inhibitor NC group and control group,the expression of miR-584-3p in miR-584-3p inhibi-tor group was down-regulated by 20 times(P <0.05).Compared with inhibitor NC group and control group,the cell proliferation ability of miR-584-3p inhibitor group showed no significant change(P >0.05),the cellular migration and invasive activity in miR-584-3p inhibitor group were up-regulated significantly(P <0.05).Conclusion The migration and invasion of glioblastoma cells can be inhibited by the over-expression of miR-584-3p,while the transfection of miR-584-3p inhibitor can enhance the migration and invasion of glioblastoma cells.
