目的:探讨4-羟基-7-氯-苯并恶唑-2-酮( TC-2)对大鼠肝星状细胞株( HSC-T6)增殖的影响。方法将HSC-T6细胞分别用不同浓度的TC-2(浓度分别为5.0、1.0、0.2、0.04、0.008 mg/ml)处理24 h,与对照组(单纯培养液)比较。甲基四唑蓝法检测细胞生长抑制情况;吖啶橙染色后荧光倒置显微镜下观察各组大鼠肝星状细胞HSC-T6的形态学变化。结果 TC-2在0.04 mg/ml 以上各浓度组对HSC-T6细胞均有明显抑制作用(P<0.05),浓度越高,抑制作用越明显,0.008 mg/ml浓度时抑制作用不明显( P>0.05);不同浓度TC-2(1.0、0.2、0.04 mg/ml)作用于HSC-T6细胞后均可见细胞体积缩小、核破裂、核浓缩、数量减少等变化,且浓度越高,变化越明显。结论 TC-2对HSC-T6细胞的增殖具有抑制作用。

목적:탐토4-간기-7-록-분병악서-2-동( TC-2)대대서간성상세포주( HSC-T6)증식적영향。방법장HSC-T6세포분별용불동농도적TC-2(농도분별위5.0、1.0、0.2、0.04、0.008 mg/ml)처리24 h,여대조조(단순배양액)비교。갑기사서람법검측세포생장억제정황;아정등염색후형광도치현미경하관찰각조대서간성상세포HSC-T6적형태학변화。결과 TC-2재0.04 mg/ml 이상각농도조대HSC-T6세포균유명현억제작용(P<0.05),농도월고,억제작용월명현,0.008 mg/ml농도시억제작용불명현( P>0.05);불동농도TC-2(1.0、0.2、0.04 mg/ml)작용우HSC-T6세포후균가견세포체적축소、핵파렬、핵농축、수량감소등변화,차농도월고,변화월명현。결론 TC-2대HSC-T6세포적증식구유억제작용。
Objective To explore the effect of 4-Hydroxy-7-chloro-2-benzoxazolone ( TC-2) on the proliferation of hepatic stellate cell line(HSC-T6) in rats.Methods HSC-T6 cells were treated with different concentrations of TC-2(5.0,1.0,0.2,0.04 and 0.008 mg/ml,respectively) for 24 hours,and were compared with the controls ( treated with simple culture medium ).The growth inhibition rates of the cells were determined by MTT assay .The morphological changes of HSC-T6 cells in each group were observed with inverted fluorenscence microscopy following acridine orange staining .Results TC-2 of the concentration of 0.04 mg/ml or higher had significant inhibition effects on HSC-T6 cells(P<0.05),the higher concentration,the more significant inhibition.TC-2 of the concentration of 0.008 mg/ml showed no significant inhibition effects(P>0.05).After HSC-T6 cells were treated by different concentrations of TC-2(1.0,0.2,0.04 mg/ml),the changes including cell shrinkage ,nuclear rupture,nuclear condensation and the decreased number of HSC cells were observed ,the higher concentration,the more significant changes.Conclusion TC-2 has the inhibitory effect on the proliferation of HSC-T6 cells.
