目的:探讨黄芪对脓毒症大鼠模型外周血单核细胞(PBMC)及支气管肺泡灌洗液(BALF)中Th1/Th2失衡的影响及意义。方法60只清洁级SD大鼠随机分为对照组[C组,早期对照组(EC组)和晚期对照组(LC)]、脓毒症组[S组,早期脓毒症组(ES)和晚期脓毒症组(LS)]及脓毒血症黄芪干预组[H组,早期干预组(EH)和晚期干预组(LH)],每组10只。采用酶联免疫吸附试验检测各组大鼠PBMC及BALF中的干扰素—γ(IFN—γ)和白介素—4(IL—4)表达水平,并计算IFN—γ/IL—4的比值。结果 S组大鼠PBMC和BALF中的IFN—γ表达均较C组和H组显著下降,且在PBMC中这种下降趋势呈时间依赖性,而IL—4表达水平显著上升,IFN—γ/IL—4表达随着时间延续进行性下调,差异均有统计学意义(P<0.01或0.05)。结论辅助性T细胞功能失衡参与了脓毒症的进展,黄芪可抑制脓毒症大鼠中的炎性反应,改善其免疫失衡。

목적:탐토황기대농독증대서모형외주혈단핵세포(PBMC)급지기관폐포관세액(BALF)중Th1/Th2실형적영향급의의。방법60지청길급SD대서수궤분위대조조[C조,조기대조조(EC조)화만기대조조(LC)]、농독증조[S조,조기농독증조(ES)화만기농독증조(LS)]급농독혈증황기간예조[H조,조기간예조(EH)화만기간예조(LH)],매조10지。채용매련면역흡부시험검측각조대서PBMC급BALF중적간우소—γ(IFN—γ)화백개소—4(IL—4)표체수평,병계산IFN—γ/IL—4적비치。결과 S조대서PBMC화BALF중적IFN—γ표체균교C조화H조현저하강,차재PBMC중저충하강추세정시간의뢰성,이IL—4표체수평현저상승,IFN—γ/IL—4표체수착시간연속진행성하조,차이균유통계학의의(P<0.01혹0.05)。결론보조성T세포공능실형삼여료농독증적진전,황기가억제농독증대서중적염성반응,개선기면역실형。
Objective To approach the effect and significance of astragalus membranaceus to imbalance of Th1/Th2 in the peripheral blood mononuclear cell(PBMC) and bronchoalveolar lavage fluid(BALF) of septic rats. Methods A total of clean 60 SD rats were randomly divided into the control group[group C including group EC and group LC] ,the sepsis group [group S in—cluding group ES and group LS] and astragalus membranaceus intervening group[group H including group EH and group LH] ,10 of each group. It adopted enzyme—linked immunosorbent assay to detect the expression level of IFN—γ and IL—4 in PBMC and BALF in the rats of each group and calculate the ratio of IFN—γ/IL—4. Results Compared to the group C and group H ,the expres—sion level of IFN—γin the PBMC and BALF of group S were significantly decreased. The downtrend in the PBMC showed time de—pendence while the IL—4 expression level were improved obviously. With the time prolonging ,the expression of IFN—γ/IL—4 in the PBMC and BALF of septic rats was dropped progressively. The difference had statistical significance (P<0.01 or 0.05). Conclu—sion The imbalance of helper T cells function participates to the development of sepsis. Astragalus membranaceus may inhibit the inflammatory response of astragalus membranaceus to improve their immunologic imbalance.
