目的:研究高脂饮食喂养诱导的胰岛素抵抗(IR)小鼠骨骼肌中磷脂酰肌醇3激酶(PI3K)表达及肌糖原含量的改变。方法将20只雄性 KM小鼠随机分为对照组(NC 组,10只)和 IR 组(10只),NC 组予常规饲料,IR 组予高脂饲料。16周时 IR 造模成功,记录小鼠体质量、空腹血糖(FBG)和空腹胰岛素(FINS),并计算稳态模型胰岛素抵抗指数(HOMA-IR);Western blot 法检测骨骼肌 PI3K 的表达,过碘酸雪夫(PAS)染色观察骨骼肌糖原含量的变化。结果与 NC 组比较,IR 组小鼠体质量、FBG、FINS 及 HOMA-IR 增高(P 均<0.05),IR 组骨骼肌中 PI3K 的表达显著降低(P <0.05);PAS 染色显示 NC 组骨骼肌细胞内紫红色着色广泛,着色颗粒较多,IR 组着色浅,着色颗粒较少。结论骨骼肌 PI3K 表达降低可能是导致肌肉 IR 产生的原因之一。

목적:연구고지음식위양유도적이도소저항(IR)소서골격기중린지선기순3격매(PI3K)표체급기당원함량적개변。방법장20지웅성 KM소서수궤분위대조조(NC 조,10지)화 IR 조(10지),NC 조여상규사료,IR 조여고지사료。16주시 IR 조모성공,기록소서체질량、공복혈당(FBG)화공복이도소(FINS),병계산은태모형이도소저항지수(HOMA-IR);Western blot 법검측골격기 PI3K 적표체,과전산설부(PAS)염색관찰골격기당원함량적변화。결과여 NC 조비교,IR 조소서체질량、FBG、FINS 급 HOMA-IR 증고(P 균<0.05),IR 조골격기중 PI3K 적표체현저강저(P <0.05);PAS 염색현시 NC 조골격기세포내자홍색착색엄범,착색과립교다,IR 조착색천,착색과립교소。결론골격기 PI3K 표체강저가능시도치기육 IR 산생적원인지일。
Objective To study the changes in phosphatidylinositol 3-kinase(PI3K)expression and glycogen content in the skeletal muscle of mice with insulin resistance(IR)fed with high fat diet. Methods 20 male KMmice were randomly divided into normal control group(NC group,n =10)and IR group(n =10).The mice in NC group were fed with routine diet and the mice in IR group were fed with high-fat diet.At the 16th week,IR models were tested successful.Record body weight,fasting blood glu-cose(FBG)and fasting insulin(FINS),then calculated Homeostasis Model Assessment for Insulin Resist-ance(HOMA-IR).The expression of PI3K in the skeletal muscle were measured by Western blotting method and the muscle glycogen content were measured by Periodic Acid-Schiff Staln(PAS).Results Compared with NC group,the body weight,FBG,FINS and HOMA-IR of IR group were higher(respective-ly P <0.05),the PI3K in the skeletal muscle of IR group decreased significantly(P <0.05).The skeletal muscle cells in NC control were colored extensively and more colored granules,while IR gruop were col-ored light and less colored granules.Conclusion The reduced expression of PI3K in skeletal muscle may be one of the reasons leading to muscle IR.
