以同期生长的梁山慈竹体细胞突变体No.29植株和实生植株(对照)为材料,对其茎秆化学成分和光合生理相关指标进行测定,并基于RNA高通量转录组测序结果,对No.29植株的纤维素、木质素等合成有关的基因差异表达进行分析。结果表明,与实生植株相比,No.29植株具有较高的株高、胸径、茎秆的鲜重和干重以及纤维素含量、纤维长度和宽度;纤维素含量比实生植株高12.89%;外方和内方纤维股明显增大;灰分含量、卷曲指数和扭结指数明显降低;但发笋量、木质素含量和细小纤维含量没有明显差异。 No.29植株中的CesA、SuSy、UDPase基因分别比实生植株上调了117%、205%、115%,转录因子MYB基因表达上调了214%,驱动蛋白基因表达上调了10倍,动力蛋白基因表达上调了760%。 No.29植株纤维素含量与纤维形态上明显的改变有其遗传基础,为No.29的进一步研究与利用奠定了基础。

The chemical components of stem and photosynthetic physiological indexes in the somatic mutant No .29 and the plant of seeds ( CK) were determined .The different expressed genes involved in the biosynthesis of cellulose and lignin from No .29 were analyzed based on RNA high-throughput sequencing , compared with those of CK .The results showed that , compared with the plant of seeds, the significant plant height, diameter at breast height, fresh weight and dry weight of stem, cellulose content, length and width of fiber were found in the mutant No.29.The cellulose content in the mutant No.29 increased 12.89%, compared with CK. The fibre strand outside the central strand and fibre strand inside the central strand and in the mutant No .29 were obviously higher than those of CK .The ash content , curl index and kink index were significantly lower than those of CK .The shooting number , lignin content and fiber fines content were insignificant , compared with CK .The expression of gene of CesA, SuSy, UDPase, transcriptional factor MYB in the mutant No .29 was up regulated by 117%, 205%, 115%, 214%, respectively .The kinesin was up regulated by 10 fold while the dynein was up regulated by 760%.The results indicated that the significant changes of cellulose content and fiber morphology have the genetic bases in the mutant No .29.It laid a foundation for further research and utilization of No .29.
