目的:评价微粒皮复合人工真皮构建的组织工程皮肤用于修复大鼠背部巨大创面的效果。方法:选取32只8周龄SD大鼠,制备鼠背巨大创面模型,并随机分为空白对照组(A组)、人工真皮组(B组)、微粒皮组(C组)及组织工程皮肤组(D组)。B、C、D组大鼠分别以打孔拉网的人工真皮、自体原位微粒皮移植物、自体原位微粒皮复合人工真皮构建的组织工程皮肤移植物修复创面,A组创面仅覆盖油纱布。观察术后第1、3、5周时的创面愈合情况;通过CK15及CD29免疫组织化学染色法观察表皮干细胞(epidermal stem cells ,ESCs)在创面愈合过程中的作用;对鼠背组织切片进行 HE染色、Masson染色后镜下观察,并结合扫描电镜下观察,了解创面真皮层的愈合质量。结果:D组大鼠背部创面的修复效果最佳,术后第5周时创面已基本愈合。免疫组织化学染色结果显示,D组大鼠ESCs逐渐向表皮层迁移并参与创面的愈合;D组真皮层的厚度大于其余3组(P<0.01),D组真皮层胶原纤维排列致密整齐。结论:微粒皮复合人工真皮构建的组织工程皮肤用于创面修复的效果优于微粒皮及人工真皮。
目的:評價微粒皮複閤人工真皮構建的組織工程皮膚用于脩複大鼠揹部巨大創麵的效果。方法:選取32隻8週齡SD大鼠,製備鼠揹巨大創麵模型,併隨機分為空白對照組(A組)、人工真皮組(B組)、微粒皮組(C組)及組織工程皮膚組(D組)。B、C、D組大鼠分彆以打孔拉網的人工真皮、自體原位微粒皮移植物、自體原位微粒皮複閤人工真皮構建的組織工程皮膚移植物脩複創麵,A組創麵僅覆蓋油紗佈。觀察術後第1、3、5週時的創麵愈閤情況;通過CK15及CD29免疫組織化學染色法觀察錶皮榦細胞(epidermal stem cells ,ESCs)在創麵愈閤過程中的作用;對鼠揹組織切片進行 HE染色、Masson染色後鏡下觀察,併結閤掃描電鏡下觀察,瞭解創麵真皮層的愈閤質量。結果:D組大鼠揹部創麵的脩複效果最佳,術後第5週時創麵已基本愈閤。免疫組織化學染色結果顯示,D組大鼠ESCs逐漸嚮錶皮層遷移併參與創麵的愈閤;D組真皮層的厚度大于其餘3組(P<0.01),D組真皮層膠原纖維排列緻密整齊。結論:微粒皮複閤人工真皮構建的組織工程皮膚用于創麵脩複的效果優于微粒皮及人工真皮。
목적:평개미립피복합인공진피구건적조직공정피부용우수복대서배부거대창면적효과。방법:선취32지8주령SD대서,제비서배거대창면모형,병수궤분위공백대조조(A조)、인공진피조(B조)、미립피조(C조)급조직공정피부조(D조)。B、C、D조대서분별이타공랍망적인공진피、자체원위미립피이식물、자체원위미립피복합인공진피구건적조직공정피부이식물수복창면,A조창면부복개유사포。관찰술후제1、3、5주시적창면유합정황;통과CK15급CD29면역조직화학염색법관찰표피간세포(epidermal stem cells ,ESCs)재창면유합과정중적작용;대서배조직절편진행 HE염색、Masson염색후경하관찰,병결합소묘전경하관찰,료해창면진피층적유합질량。결과:D조대서배부창면적수복효과최가,술후제5주시창면이기본유합。면역조직화학염색결과현시,D조대서ESCs축점향표피층천이병삼여창면적유합;D조진피층적후도대우기여3조(P<0.01),D조진피층효원섬유배렬치밀정제。결론:미립피복합인공진피구건적조직공정피부용우창면수복적효과우우미립피급인공진피。
Objective:To evaluate the effect of tissue engineering skin(TES) ,which was made of microskin combined with artificial dermis ,in the healing of huge wound on the back of SD rats .Methods:The models of huge wound on the back were made from the selected 32 SD rats at 8 weeks old .They were randomly divided into blank control group (Group A) ,artificial dermis group (Group B) ,microskin group (Group C) and TES group (Group D) .The wounds in Group B ,Group C and Group D were healed by artificial dermis with punching net ,in‐situ microskin autografts ,the TES made of autogenous in‐situ microskin combined with artificial dermis , respectively . The wounds in Group A were covered with vaseline gauze . The situations of wound healing at the 1st ,3rd and 5th week after operation were observed .The effects of ESCs during the wound healing process were observed by the CK15 and CD29 immunohistochemical stalning .The healing quality of dermis on wound was evaluated by HE stalning and Masson stalning of tissue biopsies of the back of rats and observation under scanning electronic microscopy .Results:The best healing effect was in Group D and the wounds were almost healed at the 5th week after grafting .The results of immunohistochemical stalning showed that ESCs of rats in Group D had moved to the epidermal layer gradually and took part in the wound healing .The thickness of dermal layer in Group D was larger than that in the other 3 groups(P<0 .01) and the collagen fibers in dermal layer were arranged compactly and neatly in Group D .Conclusions:The effect of the TES made of microskin combined with artificial dermis in the application of wound healing is superior to that of microskin and artificial dermis .
