目的:探讨肺纤方提取物体外干预肺纤维化微血管生成的作用及机制。方法从肺纤维化模型大鼠分离培养肺微血管内皮细胞,将其等量分为空白组、氯沙坦组、泼尼松组、肺纤方大剂量组、肺纤方中剂量组、肺纤方小剂量组。对各组的迁移细胞及微血管内皮细胞形成的血管进行计数。结果与空白组和泼尼松组相比,肺纤方大、中剂量、氯沙坦均能抑制内皮细胞的迁移(P<0.05)。氯沙坦、肺纤方各剂量均能抑制血管形成(P<0.05),其中肺纤方大剂量几乎完全抑制血管形成。结论肺纤方具有抑制肺纤维化大鼠体外培养的肺微血管内皮细胞的迁移及新生血管形成,从而发挥抗肺纤维化作用。

목적:탐토폐섬방제취물체외간예폐섬유화미혈관생성적작용급궤제。방법종폐섬유화모형대서분리배양폐미혈관내피세포,장기등량분위공백조、록사탄조、발니송조、폐섬방대제량조、폐섬방중제량조、폐섬방소제량조。대각조적천이세포급미혈관내피세포형성적혈관진행계수。결과여공백조화발니송조상비,폐섬방대、중제량、록사탄균능억제내피세포적천이(P<0.05)。록사탄、폐섬방각제량균능억제혈관형성(P<0.05),기중폐섬방대제량궤호완전억제혈관형성。결론폐섬방구유억제폐섬유화대서체외배양적폐미혈관내피세포적천이급신생혈관형성,종이발휘항폐섬유화작용。
Objective To probe into the mechanism of Feixian Formula ( FXF ) , a Chinese herbal compound, in prevention pulmonary microvascular endothelial cells ( PMVECs) migration and angiogene-sis in vitro. Methods PMVECs isolated from rat model of pulmonary fibrosis ( PF) were cultured and divided into blank group, losartan group, prednisone group, low-, mid-, and high-dose of FXF groups. Migrating cells and new vessels in each group were counted. Results High-dose and mid-dose of FXF and losartan inhibited PMVECs migration significantly ( P <0. 05 ) . High-dose, mid-dose, low-dose of FXF and losartan inhibited formation of new vessels, even high-dose of FXF completely inhibited blood vessel formation. Conclusion The potential mechanism of anti-pulmonary fibrosis of FXF in vitro is related to inhabitation of PMVECs migration and new vessels formation.
