评价乳源肽α2-f(56-57):YS的血管紧张素转化酶(ACE)活性抑制作用及降压效果,并将其应用于发酵酸奶,制备具有降压作用的发酵酸奶。采用分光光度法检测α2-f(56-57):YS对ACE活性的抑制作用,用不同剂量的α2-f(56-57):YS及添加α2-f(56-57):YS的发酵酸奶灌胃原发性高血压大鼠(SHR),评价其降血压效果。结果表明,片段α2-f(56-57):YS具有很好的ACE活性抑制作用,其IC50值为11.89μg/mL;体内检测表明,用低、中、高(1、3、9 mg/kg mb)剂量灌胃原发性高血压大鼠(SHR),均显示有降血压作用,在灌胃后4 h,其血压分别降低了18.25、31.38和36.88 mm Hg,8 h后恢复到初始血压值。添加α2-f(56-57):YS的发酵酸奶,以10 mL/kg mb的剂量灌胃SHR,在灌胃后4 h,对SHR的收缩压(SBP)降低了32.5mm Hg,8 h后恢复到初始值。因此,α2-f(56-57):YS具有降血压作用,应用于发酵酸奶中,可制备具有降压功能的酸奶。

Objective of this research was to evaluate inhibition effect of α2-f(56-57):YS, which from milkα2-casein, on ACE (angiotensin converting enzyme)activity, and its antihypertensive efficacy. And it was applied to produce yoghourt, which had antihypertensive efficacy. The inhibition effect of α2-f (56-57):YS on ACE activity was evaluated using spectrophotometric method. The antihypertensive efficacy of α2-f(56-57):YS and the yoghourt which contain α2-f(56-57):YS was evaluated by gavage SHR(pontaneously hypertensive rat) in different dose. Results showed thatα2-f(56-57):YS had good inhibition effect on ACE activity in vitro, and its median inhibitory concentration (IC50) was 11.89μg/mL. SHR was given α2-f (56-57):YS in different dose (1, 3, 9 mg/kg mb) by gavage, after given 4 hour, the blood pressure of SHR was reduced 18.25, 31.38 and 36.88 mm Hg, respectively, and it was return to the initial blood pressure values after given 8 hour. SHR was given the yoghourt of containing α2-f (56-57):YS in 10 mL/kg mb by gavage, after given 4 h, its SBP was reduced 32.5 mm Hg, and after given 8 h, the SBP return to the initial value. Therefore,α2-f (56-57):YS had antihypertensive efficacy, and applied it in yoghurt, could give the yoghourt antihypertensive efficacy.
