以沙棘籽为材料,进行了沙棘籽总黄酮的提取与纯化工艺探索.用有机溶剂法,研究了乙醇浓度、料液比、提取温度、提取时间等单因素对总黄酮提取率的影响,再以 D-100、D-500、D-7003种大孔吸附树脂为材料,筛选适宜于黄酮纯化的树脂类型.结果表明,总黄铜提取最佳工艺参数为:乙醇浓度70%,料液比1∶25(g∶mL),温度60℃,提取时间4 h/次,该条件下总黄酮提取率可达4.23%;纯化最佳工艺为:以6 mg/mL 的沙棘籽粗提液上柱,流速为2 mL/min,用80%乙醇洗脱,流速1 mL/min,在树脂与洗脱液的比例为1∶3的条件下洗脱可达到最佳纯化效果,采用该工艺可使类黄酮的纯度由0.5%提高到88.46%.

Sea buckthorn seeds were used as material,the extraction and purification processes of total flavonoids were studied in this paper.The effects of alcohol concentration,ratio of solid to liquid,extraction temperature,and extraction time were researched on the extraction ratio of total flavones.Purification of flavonoids was taken by D-100,D-500,D-700,3 macroporous adsorption resins.Results showed that the pa-rameters of extraction process were as follow:ethanol concentration 70%,ratio of solid to liquid 1∶25 (g∶mL),temperature 60 ℃ and extraction time 4 h.Under this condition the extraction ratio of the total flavones was 4.23%.The purification process was:6 mg/mL column on seed crude extracts,flow rate of 2 mL/min.With 80% alcohol as the eluent,peaking 1 mL/min,under the condition of solid-liquid ratio of 1∶3-eluting efficiency was achieved the optimum.It is suggested that purity of flavonoids in the crude prod-uct can be increased from 0.5% to 88.46% by using this technology.
