Objective To explore the mechanism of neural tube defects (NTDs)induced by cyclo-phosphamide in rats by detecting the expression of Wnt-1 and Fzd-1 .Methods Pregnant rats were random-ly divided into experimental and control groups.At 8:00AM on the 13th day of pregnancy,the experimen-tal groups were intraperitoneally inj ected with cyclophosphamide (1 5 mg/kg body weight),while the con-trol groups were injected with the same dose of normal saline.The pregnant rats were sacrificed at 8,12, 24,48 hours after injection,then the fetuses were prepared into serial paraffin sections.The expressions of Wnt-1 and Fzd-1 were detected by immunofluorescence staining.Results Compared with those of the control groups,the expressions of Wnt-1 and Fzd-1 in experimental groups were not altered at 8,12 hours after inj ection.But the optical density of Wnt-1 and Fzd-1 in experimental groups declined significantly at 24,48 hours after injection.Conclusion The conduction of the classic Wnt pathway is interfered by down-regulation of Wnt-1 and Fzd-1 ,which may be one of the important reasons for NTDs induced by cyclo-phosphamide.
