目的:观察并比较扬子鳄、家鸡及大鼠中脑酪氨酸羟化酶(Tyrosine hydroxylase,TH)和神经元型一氧化氮合酶(neuronal nitric oxide synthase,nNOS)的表达及阳性神经元的分布特征,为生物进化提供比较解剖学资料,并为扬子鳄种群遗传保护积累形态学资料。方法扬子鳄6只,家鸡和SD大鼠各15只。三种动物再各分3次进行实验,每次扬子鳄2只,家鸡和 SD大鼠各5只,取中脑组织,采用免疫组织化学方法结合黑马病理图像分析系统检测三种动物中脑 TH、nNOS的表达及阳性神经元的大小、形态与细胞平均灰度值。结果扬子鳄、家鸡、大鼠中脑内均可见不同大小和形态的 TH、nNOS 阳性神经元。三者中脑的TH、nNOS阳性神经元数量均呈现大鼠最多,家鸡其次,扬子鳄较少(三组间两两比较P<0.05),细胞平均灰度值在扬子鳄、家鸡、大鼠也呈依次减少(三组间两两比较P<0.05)。三种动物中脑内 TH、nNOS阳性神经元的大小和形态存在一定的差异。结论中脑TH、nNOS在扬子鳄、家鸡及大鼠的表达差异可能与不同物种中脑执行其相关神经系统生理功能的不同有关。

목적:관찰병비교양자악、가계급대서중뇌락안산간화매(Tyrosine hydroxylase,TH)화신경원형일양화담합매(neuronal nitric oxide synthase,nNOS)적표체급양성신경원적분포특정,위생물진화제공비교해부학자료,병위양자악충군유전보호적루형태학자료。방법양자악6지,가계화SD대서각15지。삼충동물재각분3차진행실험,매차양자악2지,가계화 SD대서각5지,취중뇌조직,채용면역조직화학방법결합흑마병리도상분석계통검측삼충동물중뇌 TH、nNOS적표체급양성신경원적대소、형태여세포평균회도치。결과양자악、가계、대서중뇌내균가견불동대소화형태적 TH、nNOS 양성신경원。삼자중뇌적TH、nNOS양성신경원수량균정현대서최다,가계기차,양자악교소(삼조간량량비교P<0.05),세포평균회도치재양자악、가계、대서야정의차감소(삼조간량량비교P<0.05)。삼충동물중뇌내 TH、nNOS양성신경원적대소화형태존재일정적차이。결론중뇌TH、nNOS재양자악、가계급대서적표체차이가능여불동물충중뇌집행기상관신경계통생리공능적불동유관。
Objective To observe and compare the expressions of tyrosine hydroxylase (TH)and neuronal nitric oxide synthase (nNOS)-positive neurons in the mesencephalon of alligator sinesis (Chinese alligators),domestic chickens and rats in order to collect comparative anatomy data for investigating of the biological evolution and accumulate morphological data for population genetic protection of Chinese alliga-tors.Methods Chinese alligators (n=6),domestic chickens (n=15)and Sprague-Dawley (SD)rats (n=15)of either sex were included and cross-grouped into three groups with 2 alligators,5 chickens and 5 rats each.Animals in the three groups were simultaneously sacrificed by electric shock.The expressions of TH and nNOS in the mesencephalon were detected by immunohistochemical assay and HemaGSM 2000P anal-ysis.Results Conglomerated or diffused distribution of TH and nNOS positive neurons were commonly seen in the mesencephalon of alligator sinesis,domestic chickens and rats;they presented with diverse cel-lular morphology of various showes,whereas the cell number of different shapes varied among the three animals.The number of TH and nNOS positive neurons in the midbrain were the most in rats,and least in Alligator sinensis.The TH and nNOS reactive neurons were primarily middle-sized in the mesencepha-lon of the three animals,and large-sized cells were relatively few in number.Nevertheless,significantly increased reactive neurons to TH and nNOS were seen in corresponding segments of the mesencephalon of rats as compared with alligator sinesis (P<0.05),and the average gray value was most in alligator sinesis and least in rats (P<0.05).Conclusion The TH and nNOS positive neurons were increased in turn in the mesencephalon of the Chinese alligators,domestic chickens and rats,and the average gray value for the cells were decreased in the same order.These findings suggest that TH and nNOS distribution differences may be associated with different neural functions area carries out in the mesencephalon of different species.
