目的:探讨地黄对去卵巢骨质疏松大鼠股骨骨密度、腰椎骨组织整合素β1 mRNA 表达的影响。方法72只雌性大鼠,随机分为假手术组、模型组、雌二醇组和地黄小、中、大剂量组。假手术组仅行假手术,其余五组行卵巢切除术。术后1 w 分别灌胃给予17β-雌二醇和地黄小、中、大剂量,连续给药3个月。测定血钙、磷和碱性磷酸酶( ALP),尿钙( u-Ca)、磷、尿脱氧吡啶酚( D-Pyr)和肌酐( Cr)。之后处死动物,取出右侧股骨,测定骨密度;取出第2腰椎,测定骨钙( b-Ca)、磷( b-P)含量;取出第4腰椎,采用实时荧光逆转录聚合酶链反应测定腰椎骨整合素β1 mRNA 表达。结果与假手术组相比,模型组血清 ALP、u-Ca、D-Pyr/Cr 显著增加,股骨密度、腰椎骨整合素β1 mRNA 表达均降低。与模型组比较,地黄中、大剂量组和雌二醇组均可使血清 ALP、u-Ca、D-Pyr/Cr 排出量降低,股骨密度、腰椎骨整合素β1 mRNA 表达均增加。结论地黄能抑制由于去卵巢雌激素缺乏引发的骨转换增强,提高骨密度,促进腰椎骨组织整合素β1 mRNA 表达,增强骨质量。

목적:탐토지황대거란소골질소송대서고골골밀도、요추골조직정합소β1 mRNA 표체적영향。방법72지자성대서,수궤분위가수술조、모형조、자이순조화지황소、중、대제량조。가수술조부행가수술,기여오조행란소절제술。술후1 w 분별관위급여17β-자이순화지황소、중、대제량,련속급약3개월。측정혈개、린화감성린산매( ALP),뇨개( u-Ca)、린、뇨탈양필정분( D-Pyr)화기항( Cr)。지후처사동물,취출우측고골,측정골밀도;취출제2요추,측정골개( b-Ca)、린( b-P)함량;취출제4요추,채용실시형광역전록취합매련반응측정요추골정합소β1 mRNA 표체。결과여가수술조상비,모형조혈청 ALP、u-Ca、D-Pyr/Cr 현저증가,고골밀도、요추골정합소β1 mRNA 표체균강저。여모형조비교,지황중、대제량조화자이순조균가사혈청 ALP、u-Ca、D-Pyr/Cr 배출량강저,고골밀도、요추골정합소β1 mRNA 표체균증가。결론지황능억제유우거란소자격소결핍인발적골전환증강,제고골밀도,촉진요추골조직정합소β1 mRNA 표체,증강골질량。
Objective To explore the influence of Radix Rehmanniae on bone mineral density ( BMD) of femur,the expression of the forth lumbar vertebrae osseous integrin beta 1(Itgβ1)mRNA in ovariectomized osteoporosis model rats .Mtehods 72 Sprague Dawley fe-male rats were randomly divided into sham ,model,17 β-estrogen treated and Radix Rehmanniae treated groups (12.5, 2.5 , 5 ml/kg).The sham group was only sham operated and the other five groups were ovariectomized respectively .One week after ovariectomy ,the rats were giv-en Radix Rehmanniae(1.25, 2.5, 5 ml/kg) and 17 β-estrogen(2 mg/kg)by intragastric administration for three months sustainedly .Serum contents of calcium(s-Ca),phosphorus(s-P),alkaline phosphate(ALP)and levels of urinary deoxypyridinoline (D-Pyr),urine contents of calcium(u-Ca),phosphorus(u-P)and creatinine(Cr)were detected.Rats were sacrificed at the end of the experiment .The thighbone was taken out to determine bone mineral density(BMD).The second lumbar vertebra was taken out to determine contents of calcium (bC-a)and phosphorus(b-P ).The forth lumbar vertebrae(LV4)was taken out to FQ-PCR for testing the expression of forth lumbar vertebra osseous inte-grin beta 1(Itgβ1)mRNA.Results Compared with those of sham group,serum ALP activity as well as u-Ca and urinary D-Pyr /Cr signifi-cantly were increased in ovariectomized rats .BMD of femur ,and the expression of the forth lumbar vertebra osseous integrin beta 1 ( Itg β1 ) mRNA was significantly decreased in model group .Compared with that of model group ,middle and high doses of Radix Rehmanniae and 17β-estrogen inhibited serum ALP activity inhibit ,and u-Ca,urinary D-Pyr/Cr reduce.BMD of femur,and the expression of the forth lumbar ver-tebra osseous integrin beta 1(Itgβ1)mRNA were increased in middle and high doses of Radix Rehmanniae groups .Conclusions Radix Re-hmanniae could inhibit the high rate of turn -over induced by ovariectomy in rats ,significantly raise BMD of the thighbone and improve the ex -pression of the forth lumbar vertebra osseous integrin beta 1( Itg β1) mRNA,and activate bone metabolism to reinforce quality of the bone .
