目的观察美洲大蠊提取液( ACE )对大鼠溃疡性结肠炎( UC )的治疗效果及对大鼠结肠黏膜表皮生长因子(EGF)、胰岛素样生长因子-1(IGF-1)表达的影响,并与地塞米松作比较。方法用2,4,6-三硝基苯磺酸(TNBS)灌肠制作大鼠UC模型,取40只Sprague Dawley大鼠,随机均分为ACE组、地塞米松组、模型对照组、正常对照组4组,每组10只。造模并干预性治疗10d后观察每组大鼠疾病活动指数(DAI)及肠黏膜损伤指数评分(CMDI),并检测结肠黏膜组织中EGF及IGF-1的表达水平。结果<br> 与模型对照组相比,ACE及地塞米松组DAI评分及CMDI评分显著降低(P<0.01);与地塞米松组相比,ACE组DAI评分较高(P<0.05),而CMDI评分差距无统计学意义(P>0.05)。与模型对照组对比,ACE组及地塞米松组的EGF表达水平升高(P<0.05), ACE组与地塞米松组IGF-1表达水平均降低(P<0.05)。结论 ACE对TNBS诱导的大鼠UC有治疗作用,其作用较地塞米松弱,上调EGF及下调IGF-1的表达,可能为其治疗机制之一。

목적관찰미주대렴제취액( ACE )대대서궤양성결장염( UC )적치료효과급대대서결장점막표피생장인자(EGF)、이도소양생장인자-1(IGF-1)표체적영향,병여지새미송작비교。방법용2,4,6-삼초기분광산(TNBS)관장제작대서UC모형,취40지Sprague Dawley대서,수궤균분위ACE조、지새미송조、모형대조조、정상대조조4조,매조10지。조모병간예성치료10d후관찰매조대서질병활동지수(DAI)급장점막손상지수평분(CMDI),병검측결장점막조직중EGF급IGF-1적표체수평。결과<br> 여모형대조조상비,ACE급지새미송조DAI평분급CMDI평분현저강저(P<0.01);여지새미송조상비,ACE조DAI평분교고(P<0.05),이CMDI평분차거무통계학의의(P>0.05)。여모형대조조대비,ACE조급지새미송조적EGF표체수평승고(P<0.05), ACE조여지새미송조IGF-1표체수평균강저(P<0.05)。결론 ACE대TNBS유도적대서UC유치료작용,기작용교지새미송약,상조EGF급하조IGF-1적표체,가능위기치료궤제지일。
Objective To observe the therapeutic effects and the expression of Epidermal Growth Factor ( EGF) and Insulin-like Growth Factors-1(IGF-1) of America Cockroach Extract(ACE)on ulcerative colitis of rats,and to compare the effects of ACE on UC with Dexamethasone .Methods Forty healthy Sprague-Dawley rats were randomly divided into normal control group , model control group , ACE group and Dexamethasone group ,with 10 rats in each group .Colitis was induced by rectal administration of TNBS in the model control group , ACE group and Dexamethasone group .Rats in the ACE group were treated with ACE ,and the Dexamethasone group were treated with Dexam-ethasone.Disease Activity Index(DAI) and Colon Macroscopic Damage Index (CMDI) were calculated,and the expressions of EGF and IGF-1 in colon tissues were detected .Results Compared with the model control ,the DAI and CMDI scores in ACE and Dexamethasone group were decreased significantly (P<0.01).The EGF expression level in ACE group were increased (P<0.05),the IGF-1 expression level in ACE group and Dexamethasone group were decreased (P<0.05).Conclusion ACE has therapeutic effect for the UC of rats ,but its effect is relatively weak when compared with dexamethasone .The increased expression of EGF and decreased expression of IGF-1 maybe one of the possible mechanisms .
