目的:观察养血消风饮对小鼠急、慢性皮炎-湿疹模型的抑制作用。方法:用二硝基氯苯分别建立小鼠耳廓慢性皮炎-湿疹模型及背部急性皮炎-湿疹模型。取60只小鼠,10只作为空白组,其余50只在耳部造成耳廓慢性皮炎-湿疹模型,分为模型组、养血消风饮低剂量组、养血消风饮中剂量组、养血消风饮高剂量组、氯雷他定组,每组10只;另取40只小鼠,10只作为空白组,其余30只在背部造成急性皮炎-湿疹模型,分为模型组、养血消风饮高剂量组、氯雷他定组,每组10只。采用灌胃给药的方法,养血消风饮低剂量组、中剂量组、高剂量组分别按每天成人等效剂量1倍、2倍、4倍给药(灌胃给药剂量分别为28.6 g·kg-1、57.2 g· kg-1、114.4 g · kg-1),氯雷他定组选用氯雷他定,按成人 4倍等效剂量给药(给药剂量为0.0052 g·kg-1),均调整灌胃给药量为0.4 mL。空白组、模型组均给予生理盐水0.4 mL灌胃。对比观察治疗前后动物模型耳肿胀度、脏器指数及组织结构的变化。结果:慢性皮炎-湿疹模型组小鼠的耳质量差值均明显高于空白组(P<0.01),养血消风饮各剂量组和氯雷他定组的耳质量差与模型组比较,均明显减小(P<0.01或P<0.05);慢性皮炎-湿疹模型组小鼠的脾指数、胸腺指数均明显升高(P<0.01);养血消风饮各剂量组和氯雷他定组均可显著下调急性、慢性皮炎-湿疹模型小鼠的脾指数及胸腺指数(P<0.01);养血消风饮可减轻二硝基氯苯造成急性皮炎-湿疹模型的表皮坏死、细胞内和细胞间水肿、真皮内炎性细胞浸润,从而减轻其对皮肤组织结构的破坏程度。结论:养血消风饮能减轻皮炎-湿疹模型小鼠耳廓的肿胀度,并能够抑制病变过程中免疫器官的肿大反应,下调胸腺指数和脾指数,从而减轻急性皮炎-湿疹小鼠病变局部组织损伤的程度。文献引用:芦源,关洪全.养血消风饮对小鼠皮炎-湿疹模型的影响[J].中医学报,2015,30(5):700-702.

Objective:To observe the inhibitory effect of Yangxue Xiaofeng Drink on mice acute and chronic dermatitis eczema model. Methods:To establish the chronic dermatitis eczema model of mouse ear and back of acute dermatitis eczema model with two nitro chlo-robenzene.From 60 mice,10 rats as the control group,the remaining 50 in the ear caused by chronic dermatitis eczema model of auri-cle,divided into model group,low dose group of Yangxue Xiaofeng Drink,middle dose group of Yangxue Xiaofeng Drink,and high dose group of Yangxue Xiaofeng Drink,loratadine group,with 10 rats in each group;another 40 small in 10 as the blank group,the remaining 30 on the back of the model of acute dermatitis and eczema,divided into the model group,high dose of Yangxue Xiaofeng drink group, loratadine group,with 10 rats in each group.By intragastric administration method of Yangxue Xiaofeng Decoction;low dose group,mid-dle dose group and high dose group respectively according to every adult equivalent dose of 1 times,2 times and 4 times of Administra-tion (gavage doses were 28.6 g·kg-1,57.2 g·kg-1,114.4 g·kg-1),loratadine group selection of loratadine,according to 4 times the adult equivalent dose (dose of 0.005 2 g·kg-1 ),adjusted by gavage dosage was 0.4 mL.The blank group and model group were given physiological saline 0.4 mL orally.In contrast to the changes observed in animal models of ear swelling degree,organ index and structure before and after the treatment.Results:The ear quality difference of chronic dermatitis eczema model group mice were signifi-cantly higher than those in the blank group (P<0.01),Chinese medicine dose group and Western medicine group in the ears of poor quality compared with the model group,were significantly decreased (P<0.01 or P<0.05 );chronic dermatitis eczema model group mice spleen index and thymus index were significantly increased(P<0.01);traditional Chinese medicine dose group and western med-icine group were significantly decreased in acute and chronic dermatitis eczema in mice spleen index and thymus index (P<0.01 );Yangxue Xiaofeng Decoction can reduce caused by acute dermatitis eczema model two nitrochlorobenzene epidermal necrosis,cellular and intercellular edema,inflammatory cell infiltration in the dermis,so as to reduce the degree of damage to the skin tissue structure. Conclusion:Yangxue Xiaofeng drink can reduce dermatitis eczema in mice auricle swelling,and swelling response suppression of im-mune organs of the pathological changes by the index of thymus and spleen,so as to reduce the acute dermatitis eczema lesion tissue damage in mice.
