目的:观察顺铂(DDP)不同疗程化疗对裸鼠人卵巢癌移植瘤的影响,并探讨其对乳腺癌耐药蛋白(BCRP)、P-糖蛋白(P-gp)和肿瘤干细胞标记物 CD133表达水平的影响。方法用人卵巢癌细胞株 SKOV3建立荷人卵巢癌裸鼠皮下移植瘤模型,24只裸鼠随机分为对照组、短疗程化疗组和足疗程化疗组,每组8只。化疗期间观察裸鼠体重和肿瘤生长状况;Western Blot 方法检测3组移植瘤组织中 BCRP、P-gp、CD133的蛋白表达水平。结果对照组移植瘤体积最大,短疗程化疗组次之,足疗程化疗组最小(P <0.05);足疗程化疗组裸鼠体重低于对照组及短疗程化疗组(P <0.01,P <0.05);足疗程化疗组 CD133、BCRP、P-gp 蛋白表达水平高于对照组及短疗程化疗组(P <0.01, P <0.05)。结论 DDP 足疗程化疗可以更好地抑制荷人卵巢癌裸鼠皮下移植瘤的生长,但残余瘤对化疗抵抗性更强,更难治疗。

목적:관찰순박(DDP)불동료정화료대라서인란소암이식류적영향,병탐토기대유선암내약단백(BCRP)、P-당단백(P-gp)화종류간세포표기물 CD133표체수평적영향。방법용인란소암세포주 SKOV3건립하인란소암라서피하이식류모형,24지라서수궤분위대조조、단료정화료조화족료정화료조,매조8지。화료기간관찰라서체중화종류생장상황;Western Blot 방법검측3조이식류조직중 BCRP、P-gp、CD133적단백표체수평。결과대조조이식류체적최대,단료정화료조차지,족료정화료조최소(P <0.05);족료정화료조라서체중저우대조조급단료정화료조(P <0.01,P <0.05);족료정화료조 CD133、BCRP、P-gp 단백표체수평고우대조조급단료정화료조(P <0.01, P <0.05)。결론 DDP 족료정화료가이경호지억제하인란소암라서피하이식류적생장,단잔여류대화료저항성경강,경난치료。
Objective To investigate the effects of different courses of chemotherapy with Cisplatin (DDP) on transplanted tumor with human ovarian cancer cells in nude mice, and on the expression levels of breast cancer resistant protein (BCRP), P-glycoprotein (P-gp) and neoplastic stem cell marker CD133. Methods Human ovarian cancer models with SKOV3 ovarian cancer of cell strains were established in 24 nude mice, and were randomly divided into con-trol group (n = 8), short-term chemotherapy group (n = 8) and full-term chemotherapy group (n = 8). The body weight and the conditions of tumor growth during the chemotherapy were observed; the expression levels of BCRP, P-gp and CD133 in tissues of transplantation tumor in the three groups were also detected using Western Blotting method. Results Tumor volume in control group was the largest one, and it was the larger one in short-term chemotherapy group, while it was the smallest one in the full-term chemotherapy group (P < 0. 05). The value of body weight in full-term chemother-apy group was significantly lower compared with those in control and short-term groups (P < 0. 01, P < 0. 05). The ex-pression levels of BCRP, P-gp and CD133 in full-term chemotherapy group were significantly increased compared with those in control and short-term chemotherapy groups (P < 0. 01, P < 0. 05). Conclusion The growth of transplanted tumor with human ovarian cancer cells in nude mice can be well inhibited with full-term chemotherapy, but the resistance of residual tumor against chemotherapy may be stronger and more difficult for treatment.
