目的:检测卵巢癌患者外周血中辅助性T细胞17(T help cell 17,Th17)/调节性T细胞(regulatory T cell, Treg)的水平并探讨其临床意义。方法:选取55例卵巢癌和60例健康对照者为研究对象,采用细胞内染色流式细胞术( Flow cytometry,FCM)、实时定量PCR(real time PCR)、酶联免疫吸附法(Enzyme linked immunosorbent assay,ELISA)法检测外周血Th17/Treg细胞百分率、核转录因子retinoid-related orphan receptor gamma-t(RORγt)/foxhead winged-helix box protein 3(Foxp3) mRNA的表达以及血浆中白细胞介素17(Interleukin-17,IL-17)/转化生长因子-β1(Transforming growth factor-β1,TGF-β1)的水平。结果:卵巢癌患者外周血中CD4+IL-17+/CD4+T细胞百分率、RORγt mRNA及IL-17水平与正常对照组相比明显升高,差异有统计学意义( P<0.05)。与正常对照组相比,卵巢癌患者外周血中CD4+Foxp3+Treg/CD4+T细胞百分率、Foxp3 mRNA及TGF-β1水平明显升高,差异有统计学意义(P<0.05)。结论:卵巢癌患者外周血中Th17/Treg细胞失衡,Th17/Treg细胞可能参与卵巢癌的发病过程。

목적:검측란소암환자외주혈중보조성T세포17(T help cell 17,Th17)/조절성T세포(regulatory T cell, Treg)적수평병탐토기림상의의。방법:선취55례란소암화60례건강대조자위연구대상,채용세포내염색류식세포술( Flow cytometry,FCM)、실시정량PCR(real time PCR)、매련면역흡부법(Enzyme linked immunosorbent assay,ELISA)법검측외주혈Th17/Treg세포백분솔、핵전록인자retinoid-related orphan receptor gamma-t(RORγt)/foxhead winged-helix box protein 3(Foxp3) mRNA적표체이급혈장중백세포개소17(Interleukin-17,IL-17)/전화생장인자-β1(Transforming growth factor-β1,TGF-β1)적수평。결과:란소암환자외주혈중CD4+IL-17+/CD4+T세포백분솔、RORγt mRNA급IL-17수평여정상대조조상비명현승고,차이유통계학의의( P<0.05)。여정상대조조상비,란소암환자외주혈중CD4+Foxp3+Treg/CD4+T세포백분솔、Foxp3 mRNA급TGF-β1수평명현승고,차이유통계학의의(P<0.05)。결론:란소암환자외주혈중Th17/Treg세포실형,Th17/Treg세포가능삼여란소암적발병과정。
Objective:To detect the levels of T help cell 17 (Th17)/regulatory T cell (Treg) in the peripheral blood of patients with ovarian cancer and explore their clinical significances.Methods:55 patients with ovarian cancer and 60 normal controls were enrolled.The percentage of Th17/Treg cells were detected by flow cytometry (FCM).The expression of retinoid-related orphan receptor gamma-t ( RORγt)/foxhead winged-helix box protein 3 ( Foxp3) mRNA was measured by real time PCR and the plasma levels of IL-17/TGF-β1 were detected by enzyme linked immunosorbent assay (ELISA).Results: The percentage of CD4+IL-17+/CD4+T cells,the mRNA expression of RORγt and the plasma levels of IL-17 increased significantly in patients with ovarian cancer than in normal controls (P<0.05).Compared with normal controls,the percentage of CD4+Foxp3+Treg/CD4+T cells,the mRNA expression of Foxp3 and the plasma levels of TGF-β1 increased significantly in patients with ovarian cancer ( P<0.05).Conclusion:The imbalance of Th17/Treg cells may exist in the peripheral blood of patients with ovarian cancer.Th17/Treg cells may participate the pathology of ovarian cancer.
