建立了测定鱼体血浆和肌肉组织中没食子酸含量的高效液相色谱--紫外法。该方法的流动相为:0.1%甲酸水溶液∶甲醇(V/V,85∶15);流速:1.3mL/min;检测波长:272nm;色谱柱:Waters XTrra C18(4.6mm×250mm,5μm)。该方法的检出限为0.01μg/mL,平均回收率为85%~110%。线性范围为0.01μg/mL~50μg/mL。本实验方法简便可靠、准确、重复性好。

A method of high performance liquid chromatography using UV detector(HPLC- UV)was developed for analysis of gallic acid residues in plasma, and muscle of fish. The tissue samples were extracted and purified by solution prior to detection. The methanol and water containing 0.1% formic acid(15:85)was used as the mobile phase at a rate of 1.3 mL/min under a chromatographic column of Waters XTrra C18(4.6 mm x 250 mm,5μm)in the method, with the limit of 0.01μg/mL, the average recovery from 85 to 110% and the linear range from 0.01μg/mL to 50μg/mL. Test revealed that this method was proved to be easy and simple processing.
