采用中心组合设计(CCD)响应面法(RSM )优化α淀粉酶抑制剂筛选条件。在单因素试验基础上,对影响α淀粉酶抑制剂活性的4个因素即淀粉溶液浓度、保温温度、DNS用量、沸水时间进行优化,根据回归方程最终确定α淀粉酶溶液质量浓度1 mg/mL ,淀粉溶液质量浓度17 mg/mL ,保温温度46℃,保温时间15 min ,DNS用量2 mL ,沸水时间6 min 。经试验验证结果稳定可靠,可用于α淀粉酶抑制剂筛选。

채용중심조합설계(CCD)향응면법(RSM )우화α정분매억제제사선조건。재단인소시험기출상,대영향α정분매억제제활성적4개인소즉정분용액농도、보온온도、DNS용량、비수시간진행우화,근거회귀방정최종학정α정분매용액질량농도1 mg/mL ,정분용액질량농도17 mg/mL ,보온온도46℃,보온시간15 min ,DNS용량2 mL ,비수시간6 min 。경시험험증결과은정가고,가용우α정분매억제제사선。
Central Composite Design (CCD) coupled with Response Surface Method (RSM ) has been per‐formed to optimize α‐amylase inhibitor screening conditions .On the basis of single factor experiments , four factors includes starch solution concentration ,heat preservation temperature ,dosage of DNS ,boiling time that affect α‐amylase activity have been optimized .Results show as follow s :α‐amylase concentration 1mg/mL ,starch solution concentration 17 mg/mL ,preservation temperature 46 ℃ ,holding time 15min , dosage of DNS 2 mL ,boiling time 6 min .The result is stable and reliable and can be used for α‐amylase in‐hibitor screening .
