应用Trizol法从山羊子宫内膜中提取RNA,根据已获得的山羊IFN基因序列设计并合成引物,采用RT-PCR技术进行IFN基因的扩增。结果表明,所提取的RNA经琼脂糖凝胶电泳呈现出28S rRNA、18S rRNA 2条清晰的条带,其OD260/OD280值为1.91,RT-PCR得到的条带与目的片断大小一致,说明所提取的RNA纯度高、无降解,可进一步用于干扰素的分子生物学研究。

응용Trizol법종산양자궁내막중제취RNA,근거이획득적산양IFN기인서렬설계병합성인물,채용RT-PCR기술진행IFN기인적확증。결과표명,소제취적RNA경경지당응효전영정현출28S rRNA、18S rRNA 2조청석적조대,기OD260/OD280치위1.91,RT-PCR득도적조대여목적편단대소일치,설명소제취적RNA순도고、무강해,가진일보용우간우소적분자생물학연구。
The total RNA was extracted from the endometrium of goat by Trizol method. IFN gene was amplificated by RT-PCR with the primers which had been designed according to the obtained goat IFN gene sequence and synthesized.The extracted RNA re?sults showed two clear bands of 28S rRNA and 18S rRNA on the map of electrophoresis.The relative value of OD260/OD280 was 1.91. The obtained band of RT-PCR was in accordance with the objective fragment.These results displayed the extracted RNA was pure and unbroken,and could be used for subsequent molecularbiologystudy.
