目的:探讨CD200减轻帕金森病(PD )病变区域炎症反应的作用。方法制备1‐甲基‐4‐苯基‐1,2,3,6‐四氢吡啶(MPTP)小鼠PD模型,取脑组织。检测CD200及其受体CD200R的表达和ATP敏感性钾通道(KATP通道)不同亚基在小胶质细胞(MG)的表达;检测MG的细胞因子;分析外源性重组CD200对KATP通道释放的影响。结果 PD模型小鼠CD200及CD200R的表达下调, M G的IFN‐γ、T N F‐α和IL‐1β的释放增多。外源性重组CD200能够抑制M G中A T P的释放,M G中KATP通道的开放增加,1‐甲基‐4‐苯基‐吡啶离子(Mpp+)引起的MG激活及炎症因子和氧化因子的释放受到抑制。结论CD200通过抑制MG内ATP的水平,促进MG中KATP通道的开放,抑制MG的活化及相关炎症因子的释放,减轻黑质纹状体区域的炎症反应,从而起到了保护多巴胺神经元的作用。

목적:탐토CD200감경파금삼병(PD )병변구역염증반응적작용。방법제비1‐갑기‐4‐분기‐1,2,3,6‐사경필정(MPTP)소서PD모형,취뇌조직。검측CD200급기수체CD200R적표체화ATP민감성갑통도(KATP통도)불동아기재소효질세포(MG)적표체;검측MG적세포인자;분석외원성중조CD200대KATP통도석방적영향。결과 PD모형소서CD200급CD200R적표체하조, M G적IFN‐γ、T N F‐α화IL‐1β적석방증다。외원성중조CD200능구억제M G중A T P적석방,M G중KATP통도적개방증가,1‐갑기‐4‐분기‐필정리자(Mpp+)인기적MG격활급염증인자화양화인자적석방수도억제。결론CD200통과억제MG내ATP적수평,촉진MG중KATP통도적개방,억제MG적활화급상관염증인자적석방,감경흑질문상체구역적염증반응,종이기도료보호다파알신경원적작용。
Objective To explore the role of CD200 in attenuating the inflammatory response in the substantia nigra area of mouse model with Parkinson′s disease(PD) .Methods PD model of mice was established by MPTP injection and brain tissues were taken for the examinations .The expressions of CD200 and its receptor (CD200R) were detected .The expressions of ATP‐sensitive potassium channel and cytokines in the subunits were determined .The effect of exogenous recombinant CD200 on the opening of ATP‐sensitive potassium channel in vitro was analyzed .Results The expressions of CD200 ,CD200R ,and the release of IFN‐γ,TNF‐αand IL‐1βin the subunits were all decreased in PD mice .The exogenous recombinant CD200 inhibited the ATP release and promoted the opening of ATP‐sensitive potassium channel in microglia .Mpp +‐induced microglia activation and release of inflammatory factors and oxygen free radicals were depressed .Conclusion CD200 might protect the dopaminergic neuron by attenuating neuroinflammation in the substantia nigra area via reducing ATP release ,promoting the opening of ATP‐sensitive potassium channel ,inhibiting microglia activation and release of inflammatory factors and oxygen free radicals .
