目的:目的探讨选择性环氧化酶-2(COX-2)抑制剂Celebrex对人卵巢癌细胞HO-8910的抑制作用及其机制。方法选用HO-8910体外培养,应用四甲基偶氮唑蓝( MTT)法、逆转录聚合酶链反应和酶联免疫吸附试验检测Celebrex对HO-8910细胞生长以及COX-2 mRNA、前列腺素E2(PGE2)和白介素-6(IL-6)蛋白表达的影响。结果 Celebrex对HO-8910细胞的生长具有抑制作用,其效应具有时间-剂量依赖性,在达到一定作用时间和剂量时能明显降低PGE2和IL-6蛋白的表达,但不影响COX-2 mRNA的表达。结论 Celebrex可能主要是通过抑制COX-2活性及PGE2和IL-6等的表达而对卵巢癌发挥抑制效应。

목적:목적탐토선택성배양화매-2(COX-2)억제제Celebrex대인란소암세포HO-8910적억제작용급기궤제。방법선용HO-8910체외배양,응용사갑기우담서람( MTT)법、역전록취합매련반응화매련면역흡부시험검측Celebrex대HO-8910세포생장이급COX-2 mRNA、전렬선소E2(PGE2)화백개소-6(IL-6)단백표체적영향。결과 Celebrex대HO-8910세포적생장구유억제작용,기효응구유시간-제량의뢰성,재체도일정작용시간화제량시능명현강저PGE2화IL-6단백적표체,단불영향COX-2 mRNA적표체。결론 Celebrex가능주요시통과억제COX-2활성급PGE2화IL-6등적표체이대란소암발휘억제효응。
Objective To study the inhibitory action and mechanism of selective cyclooxygeness-2(COX-2)in-hibitor,celebrex on ovarian cancer HO-8910 cells. Methods Methyl thiazolyl tetrazolium( MTT)assay,reverse transcription polymerase chain reaction and enzyme-linked immunosorbent assay were used to detect the effects of Celebrex on cell growth,the expression of COX-2 mRNA and the protein expressions of prostaglandin E2( PGE2 ) and interleukin-6(IL-6)in HO-8910 cells in vitro. Results Celebrex in a certain time and dose range had an in-hibitory action on the growth of HO-8910 cells. Celebrex decreased significantly the expressions of PGE2 and IL-6 protein,but had no effects on the expression of COX-2 mRNA. Conclusion Celebrex exerts a growth inhibition on ovarian cancer cells mainly by inhibiting the COX-2 protein and IL-6 plus PGE2.
