目的:探析并改进替加环素的合成工艺,为提高其应用价值提供依据。方法以米诺环素作为原料,经过硝化、还原、成盐酸盐、亲核取代反应等合成制得替加环素。结果替加环素总收率为42.2%,纯度为99.9%,差向异构体<0.1%。结论本研究优化了替加环素合成工艺,且得到高质量产品,为工业化生产提供了重要依据。

Objective To analyze and improve the synthetic process of Tigecycline and provide the basis for enhancing its application value. Methods With Minocycline as raw material,Tigecycline was synthetized through nitration,deoxidation,hydrochloride synthesis,nucleophilic substitution,etc. Results The overal yield of Tigecycline was 42.2%,its purity was 99.9%,and its epimer was less than 0.1%. Conclusion This research optimizes the synthetic process of Tigecycline so as to obtain high-quality products,providing an important basis for the industrialized production of Tigecycline.
