目的:探讨脱甲基化药物5-氮-2ˊ-脱氧胞苷(DAC)对前列腺癌细胞的生长抑制效果。方法利用WST -1法测定 DAC 对前列腺癌细胞株的增殖影响作用,流式细胞仪检测 DAC 在诱导细胞凋亡与细胞周期捕获中的效果,同时应用半胱天冬酶(Caspase)活性分析与增殖细胞核抗原(PCNA)表达来进一步阐明 DAC 的作用机制。结果DAC 同时对激素依赖型及激素非依赖性型前列腺癌细胞株具有显著的生长抑制效果,DAC 的作用机制主要是通过诱导G2/ M 期的细胞捕获实现,而不是通过凋亡诱导。结论脱甲基化药物 DAC 可抑制前列腺癌细胞的生长,将来可能成为治疗前列腺癌并改善预后的一种新型药物。

목적:탐토탈갑기화약물5-담-2ˊ-탈양포감(DAC)대전렬선암세포적생장억제효과。방법이용WST -1법측정 DAC 대전렬선암세포주적증식영향작용,류식세포의검측 DAC 재유도세포조망여세포주기포획중적효과,동시응용반광천동매(Caspase)활성분석여증식세포핵항원(PCNA)표체래진일보천명 DAC 적작용궤제。결과DAC 동시대격소의뢰형급격소비의뢰성형전렬선암세포주구유현저적생장억제효과,DAC 적작용궤제주요시통과유도G2/ M 기적세포포획실현,이불시통과조망유도。결론탈갑기화약물 DAC 가억제전렬선암세포적생장,장래가능성위치료전렬선암병개선예후적일충신형약물。
Objective To explore the effect of 5 - aza - 2ˊ- deoxycytidine(DAC),a DNA methyltransferase inhibitor,on the growth of prostate carcinoma(PC)cell lines. Methods The cytotoxicity of DAC on PC cell lines had been measured by using WST - 1 assay. Its effect on induction of apoptosis and arrest of cell cycle had been analyzed by flow cytometric analysis. Caspase activity and PCNA expression were also ap-plied to clarify the mechanism of DAC against PC. Results The suppression of growth had been confirmed by DAC in test for PC cell lines,while the inhibition of proliferation by DAC was through inducing arrest of G2 / M cell cycle but not by inducing apoptosis. Conclusion DAC has the effect of suppression on growth of PC cell lines and it may be a new strategy to treat prostate malignancy in the future.
