目的:探讨肿瘤坏死因子( TNF-β)G252A基因在子宫内膜异位症发病机制中作用。方法:80例经腹腔镜和病理确诊的内异症( Ems)患者为Ems组,80例同期需采用腹腔镜排除子宫内膜异位症的健康妇女为对照组,取两组妇女静脉血,采用限制性片段长度多态性聚合酶链反应( PCR-RFLP)技术检测TNF-β G252A基因的基因频率,酶联免疫( ELISA)法测定血清TNF-β的含量,分析子宫内膜异位症与TNFβ G252A基因之间的关系。结果:Ems组血清TNF-β的含量明显高于对照组( P<0.05),且Ems组血清中TNF-β的含量与EmT的分期呈正相关( r=0.8532);Ems组的A/A 基因型及G/A 基因型频率均明显高于对照组,差异有统计学意义( P<0.05);A/A基因型患Ems的危险度是G/G基因型的3.5倍。结论:Ems患者血清中TNF-β的含量与EmT的分期呈正相关,A/A基因型是患Ems的危险基因型。

목적:탐토종류배사인자( TNF-β)G252A기인재자궁내막이위증발병궤제중작용。방법:80례경복강경화병리학진적내이증( Ems)환자위Ems조,80례동기수채용복강경배제자궁내막이위증적건강부녀위대조조,취량조부녀정맥혈,채용한제성편단장도다태성취합매련반응( PCR-RFLP)기술검측TNF-β G252A기인적기인빈솔,매련면역( ELISA)법측정혈청TNF-β적함량,분석자궁내막이위증여TNFβ G252A기인지간적관계。결과:Ems조혈청TNF-β적함량명현고우대조조( P<0.05),차Ems조혈청중TNF-β적함량여EmT적분기정정상관( r=0.8532);Ems조적A/A 기인형급G/A 기인형빈솔균명현고우대조조,차이유통계학의의( P<0.05);A/A기인형환Ems적위험도시G/G기인형적3.5배。결론:Ems환자혈청중TNF-β적함량여EmT적분기정정상관,A/A기인형시환Ems적위험기인형。
Objective:To investigate the role of TNF-βG252A gene in pathogenesis of endometriosis ( Ems ). Methods:Eighty Ems patients diagnosed by laparoscopy and histopathologic examination served as Ems group and 80 healthy women served as control group. PCR-RFLP was used to detect gene frequency of TNF-βG252A gene,and ELISA was applied to check the serum content of TNF-β. The relationship of Ems and TNF-β G252A gene was analyzed. Result:The serum level of TNF-β in Ems group was higher than that in control group( P<0 . 05 ),and the serum level of TNF-β in Ems group was positively associated with EmT period. A/A and G/A genotypic frequencies in Ems group were significantly higher than those in control group. The risk of Ems of A/A genotype was 3. 5 times of G/G genotype. Conclusions:The serum level of TNF-βin Ems patients is positively associated with EmT period,and A/A genotype is the risk genotype of Ems.
