目的::研究刺五加皂苷对高脂高糖饮食大鼠学习记忆能力的影响和机制。方法:将24只大鼠随机分为对照组、高脂高糖饮食组和刺五加皂苷干预组,每组各8只。通过高脂高糖饮食建立大鼠模型,刺五加皂苷腹腔注射进行干预,Y 迷宫实验检测大鼠学习记忆能力,电生理实验检测海马 CA1区长时程增强,Westernblot 方法检测海马 GSK-3β和磷酸化 GSK-3β的表达。结果:刺五加皂苷组大鼠 Y 迷宫学习和记忆成绩明显高于高脂高糖饮食组,刺五加皂苷能增强高脂高糖饮食大鼠海马 CA1区长时程(long-term potentiation,LTP),并且抑制海马脑组织中 GSK-3β磷酸化。结论:刺五加皂苷能改善高脂高糖饮食大鼠学习记忆能力和降低突触的可塑性,其机制与抑制海马 GSK-3β磷酸化有关。

목적::연구자오가조감대고지고당음식대서학습기억능력적영향화궤제。방법:장24지대서수궤분위대조조、고지고당음식조화자오가조감간예조,매조각8지。통과고지고당음식건립대서모형,자오가조감복강주사진행간예,Y 미궁실험검측대서학습기억능력,전생리실험검측해마 CA1구장시정증강,Westernblot 방법검측해마 GSK-3β화린산화 GSK-3β적표체。결과:자오가조감조대서 Y 미궁학습화기억성적명현고우고지고당음식조,자오가조감능증강고지고당음식대서해마 CA1구장시정(long-term potentiation,LTP),병차억제해마뇌조직중 GSK-3β린산화。결론:자오가조감능개선고지고당음식대서학습기억능력화강저돌촉적가소성,기궤제여억제해마 GSK-3β린산화유관。
Objective: To observe effects of Acanthopanax senticosus saponins (ASS) on learning and memory abilities of high fat/ high sugar diet (HF/ HS) rats and discuss its mechanism. Methods: 24 male SD rats were randomly divided into control group, HF/ HS group and ASS group with 8 rats in each group. The rats in HF/ HS group and ASS group were fed with high fat/ high sugar diet for 60 days to establish rat model. Then, the rats in ASS group were treated with ASS solution through intraperitoneal injection. After that, the learning and memory abilities were detected with Y maze test. The electrophysiological method was used to observe the long-term potentiation (LTP) in hippocampal CA1 area, and Western blot was used for expression of glycogen synthase kinase-3β (GSK-3β) and phosphorylated GSK-3β (p-GSK-3β) in the hippocampal tissue. Results: The learning and memory abilities of ASS group were significantly better than those of HF/ HS group through the Y maze test. ASS could improve the LTP in hippocampal CA1 area of HF/ HS rats, and inhibit the phosphorylation of GSK-3β in the hippocampus. Conclusions: ASS can improve learning and memory a-bilities and reduce synaptic plasticity of high fat/ high sugar diet rats, and these effects could be associated with the inhibition of phos-phorylation of GSK-3β.
