人工合成苯巴比妥完全抗原并进行鉴定。对苯巴比妥结构进行修饰,合成带氨基基团的苯巴比妥半抗原,经重氮法与 BSA 和 OVA 制备苯巴比妥人工抗原。通过 TLC、质谱核磁共振对苯巴比妥半抗原进行鉴定,并采用紫外扫描对人工抗原进行结构鉴定,通过动物免疫对人工抗原进行生物学鉴定,使用 ELISA 对得到的抗血清进行鉴定分析。结果表明:苯巴比妥半抗原合成成功,苯巴比妥与 BSA,OVA 偶联成功,经计算其偶联比分别为231和10.91.动物免疫表明,免疫的5只小鼠的效价均在104以上,且4号小鼠的抗血清抗苯巴比妥 IC50为175.19 ng·mL-1。此实验成功合成了苯巴比妥人工抗原,为进一步制备苯巴比妥的特异性抗体与建立苯巴比妥免疫检测方法奠定了基础。
人工閤成苯巴比妥完全抗原併進行鑒定。對苯巴比妥結構進行脩飾,閤成帶氨基基糰的苯巴比妥半抗原,經重氮法與 BSA 和 OVA 製備苯巴比妥人工抗原。通過 TLC、質譜覈磁共振對苯巴比妥半抗原進行鑒定,併採用紫外掃描對人工抗原進行結構鑒定,通過動物免疫對人工抗原進行生物學鑒定,使用 ELISA 對得到的抗血清進行鑒定分析。結果錶明:苯巴比妥半抗原閤成成功,苯巴比妥與 BSA,OVA 偶聯成功,經計算其偶聯比分彆為231和10.91.動物免疫錶明,免疫的5隻小鼠的效價均在104以上,且4號小鼠的抗血清抗苯巴比妥 IC50為175.19 ng·mL-1。此實驗成功閤成瞭苯巴比妥人工抗原,為進一步製備苯巴比妥的特異性抗體與建立苯巴比妥免疫檢測方法奠定瞭基礎。

The current study aimed to synthesize the complete antigen of phenobarbital.Artificial phenobar-bital hapten with the active group amino group was designed and synthesized by chemical modification and linked to BSA and OVA through the diazotization method.Phenobarbital hapten was identified by TLC、MS.The structure of artificial phenobarbital antigen was identified by ultraviolet scanning.Moreover,the biological identification of artificial phenobarbital antigen was done by mice immunization.Polyclonal anti-serum was obtained,identified and analyzed by ELISA.The results showed that the synthesis of phenobar-bital hapten was successful.The phenobarbital was coupled with BSA and OVA,and the conjugation ratio was about 231 and 10.91.The animal immunization results showed that titer against phenobarbital of five mice were above 104 and IC50 of NO.4 mice was 175.19ng/mL.Artificial Phenobarbital antigen were synthesized and confirmed in the study.Therefore,it is possible to establish a base of the generation specif-ic antibody and development of immunoassay of phenobarbital.
