目的:比较乳腺癌患者自体细胞因子诱导的杀伤细胞(cytokine -induced killer cells ,CIK)与异体(健康成人)CIK细胞的抗肿瘤效应。方法应用常规方法分别自乳腺癌患者及健康成人外周血单个核细胞诱导CIK 细胞,用流式细胞术分析免疫表型,ELISA 法测定其分泌干扰素-γ(IFN -γ)水平。同时应用乳酸脱氢酶法(LDH)检测两组CIK细胞对人乳腺癌细胞株SKBR-3的杀伤活性。结果异体CIK细胞增殖能力明显高于自体CIK细胞( P<0.01),CD3+CD8+、CD3+CD56+细胞比率较相同条件下自体CIK细胞组显著增多( P<0.05)。培养第15日,异体CIK细胞分泌IFN -γ水平高于自体CIK细胞( P<0.05),且对人乳腺癌细胞株SKBR-3细胞的杀伤率显著高于自体CIK细胞( P<0.01)。结论健康成人CIK 细胞比乳腺癌患者的自体CIK 细胞有更强的抗肿瘤效应。

Objective To compare the anti -tumor immune response of autologous cytokine - induced killer ( CIK) cells which induced from breast cancer patients with that of allogeneic CIK cells which induced from healthy adults .Methods Using conventional methods induce CIK cells from two kind of peripheral blood mononuclear cells (the breast cancer patients and health adult ) respectively .The killing activity was detected by LDH assay ;immunophenotype changes were analyzed by flow cytometry ;the IFN -γlevel of the cultured su-pernatants were detected by ELISA .Results The results showed that the proliferation capability of allogeneic CIK cells was significantly higher than that of autologous CIK cells .Compared with autologous CIK cells ,allo-geneic CIK cells significantly enhanced the anti -tumor activity to killing SKBR -3 cells and IFN -γsecretion , increasing the ratio of CD3 + CD5 6 + cells and CD3 + CD8 + cells .Conclusion CIK cells induced from healthy adults have a stronger anti -tumor effect than which induced from breast cancer patients .The results can provide experimental evidence for clinical application of CIK cells .
