目的:观察MK-801对脑缺血大鼠神经功能缺损及脑神经生长因子( NGF)表达的影响,探讨MK-801治疗脑缺血的机制。方法采用线栓法制做脑缺血大鼠模型,将大鼠分为三组:假手术组、模型组和药物组,每组各24只。药物组腹腔注射MK-801(0.6mg/kg/d),其余注射同容积生理盐水。于注射后3天、7天、14天每组各取8只进行神经功能缺损评分及处死取脑,应用酶联免疫吸附试验( ELISA法)测定NGF含量。结果神经功能缺损评分在第3天模型组和药物组没有显著差异,第7天、第14天药物组较模型组有显著差异( P <0.01);NGF表达在第3天模型组和药物组没有显著差异,但与假手术组有显著差异( P <0.001或P <0.05),第7天、第14天药物组较模型组有显著差异( P <0.001或P <0.05)。结论MK-801通过提高脑内NGF的表达,改善运动功能状况可能是其治疗缺血性脑血管疾病的机制之一。

목적:관찰MK-801대뇌결혈대서신경공능결손급뇌신경생장인자( NGF)표체적영향,탐토MK-801치료뇌결혈적궤제。방법채용선전법제주뇌결혈대서모형,장대서분위삼조:가수술조、모형조화약물조,매조각24지。약물조복강주사MK-801(0.6mg/kg/d),기여주사동용적생리염수。우주사후3천、7천、14천매조각취8지진행신경공능결손평분급처사취뇌,응용매련면역흡부시험( ELISA법)측정NGF함량。결과신경공능결손평분재제3천모형조화약물조몰유현저차이,제7천、제14천약물조교모형조유현저차이( P <0.01);NGF표체재제3천모형조화약물조몰유현저차이,단여가수술조유현저차이( P <0.001혹P <0.05),제7천、제14천약물조교모형조유현저차이( P <0.001혹P <0.05)。결론MK-801통과제고뇌내NGF적표체,개선운동공능상황가능시기치료결혈성뇌혈관질병적궤제지일。
Objective To observe the effects of MK -801 on neurologic impairment and brain nerve growth factor ( NGF) ex-pression in cerebral ischemia rats ,moreover explore the mechanism of MK -801 treating cerebral ischemia.Methods The rats model of cerebral ischemia induced by line switch method were randomly divided into three groups : sham operation group、model group and drug group(n=24 each).Drug group underwent intraperitoneal injection of MK -801 (0.6mg/kg/d),other two groups were given e-quivalent saline at the same time.At 3rd、7th、14th day after injection,8rats were taken from each group to implement neurologic impair-ment score and decapitation ,the content of NGF determined by using enzyme linked immunosorbent assay (ELISA).Results Neuro-logic impairment score had no obvious differences bewteen model group and drug group at 3rd day,but drug group had obvious differ-ences compared with model group at 7th、14th day( P <0.01).The expression of NGF had no obvious differences between model group and drug group,but there are obvious differences compared with sham operation group ( P <0.001, P <0.05) at 3rd day,drug model show obvious differences compared with model group at 7th、14th day( P <0.001, P <0.05).Conclusion MK-801 could enhance the expression of brain NGF and improve motor function ,this may be one of the mechanisms by which MK -801 could treat ischemic cerebrovascular disease.
