利用化学发光免疫技术测定了3头隐睾克隆猪和3头正常猪不同阶段(2—6月龄)的睾酮(T)和雌二醇(E2)激素的水平;运用相对荧光定量 PCR 技术检测 AR 和 ER 基因在睾丸组织中的 mRNA 表达量。结果表明:正常猪血清 T 和 E2浓度从2月龄到6月龄呈上升趋势,隐睾克隆猪血清 E2浓度变化趋势与对照组相似, T 变化趋势为先缓慢升高然后降低,5月龄达到最高[(1.98±1.06)ng?mL],且各阶段血清浓度均显著低于正常猪;ER 基因在克隆猪睾丸中表达量高于对照组,但差异不显著,AR 基因表达量则反之,且差异显著。血清 T 的低水平和睾丸 AR 基因 mRNA 低表达均可能是引起克隆猪发生隐睾的因素。

In this study,the serum levels of testosterone(T)and estrogen(E2 )were detected by fluorescent immunocytometry technology in 3 cloned pigs with cryptorchidism and 3 normal pigs at different stages(2—6 months old).RT-PCR technique was used to analysis the mRNA expressions of AR and ER gene in testicular tis-sue.The results showed that the serum levels of T and E2 in normal pigs increased with their growth and develop-ment,and the trend of E2 in the cloned pigs was similar with those of normal pigs.The concentration of T in-creased and reached the highest in 5 months old[(1.98 ±1.06)ng?mL],then decreased.Serum levels of Tin cloned pigs were much lower than those of normal pigs.The expression levels of ER gene in cloned pigs were higher than that of normal pigs,but there were no significant differences between them.On the contrary,the ex-pression levels of AR gene in cloned pigs were much lower than those of normal pigs.Results indicated that the lower serum level of T and the abnormal expression of AR gene may be the important factors for the occurrence of cryptorchidism in cloned pigs.
