目的:通过比较卵巢功能减退患者与正常人血浆 Mir-21表达差异及颗粒细胞凋亡情况,探讨 Mir-21对卵巢功能的影响,为研究卵巢功能减退的发病机制提供理论依据。方法根据基础卵泡刺激素(FSH)值,将研究对象分为卵巢功能减退组(FSH >10 U /L)20例和正常组(FSH <8 U /L)23例,根据数据库和既往研究,通过实时定量 PCR的方法检测外周血中与细胞凋亡相关的 Mir-21表达量,同时比较颗粒细胞的凋亡情况。结果卵巢功能减退组外周血中 Mir-21表达低于正常组(P <0.05),颗粒细胞凋亡率明显高于正常组(P <0.05)。hoechst 33258染色发现卵巢功能减退组颗粒细胞亮染,细胞核固缩,核内出现细小不规则颗粒状凋亡小体。结论卵巢功能减退患者较正常人外周血中 Mir-21表达下调,可能会导致颗粒细胞凋亡增加,卵母细胞发育异常,导致卵巢功能减退。

목적:통과비교란소공능감퇴환자여정상인혈장 Mir-21표체차이급과립세포조망정황,탐토 Mir-21대란소공능적영향,위연구란소공능감퇴적발병궤제제공이론의거。방법근거기출란포자격소(FSH)치,장연구대상분위란소공능감퇴조(FSH >10 U /L)20례화정상조(FSH <8 U /L)23례,근거수거고화기왕연구,통과실시정량 PCR적방법검측외주혈중여세포조망상관적 Mir-21표체량,동시비교과립세포적조망정황。결과란소공능감퇴조외주혈중 Mir-21표체저우정상조(P <0.05),과립세포조망솔명현고우정상조(P <0.05)。hoechst 33258염색발현란소공능감퇴조과립세포량염,세포핵고축,핵내출현세소불규칙과립상조망소체。결론란소공능감퇴환자교정상인외주혈중 Mir-21표체하조,가능회도치과립세포조망증가,란모세포발육이상,도치란소공능감퇴。
Objective To compare the differences of plasma Mir-21 expression and granular cell apoptosis between patients with hypovaria and healthy people,and to explore the influence of Mir-21 on ovarian function in order to provide theoretical basis for pathogenesis of ovarian function decline.Methods According to basic FSH,the subjects were divided into hypovaria group (FSH >10 U /L,n =20)and control group (FSH < 8 U /L,n =23).According to the database and previous study,the expression levels of Mir-21 were detected by Real-time quantitative PCR,and the apoptosis rate of granulosa cells was compared between two groups.Results The expression levels of Mir-21 in hypovaria group were significantly lower than those in control group (P <0.05),however,the apoptosis rate of granular cell was significantly higher than that of control group (P <0.05 ).The hoechst 33258 staining showed that the granulosa cells in hypovaria group were brightly stained,with karyopyknosis and irregular granular apoptotic body in cell nucleus.Conclusion The expression levels of Mir-21 in peripheral blood of patients with hypovaria are down-regulated,as compared with those in healthy people,which may result in the increase of granular cell apoptosis and abnormal development of oocyte,so as to result in hypovaria.
